2014—2015年黑龙江省哈尔滨地区犬冠状病毒M基因分子流行病学调查 被引量:4

摘要 为调查黑龙江省哈尔滨地区犬冠状病毒(CCo V)的流行情况,本研究通过RT-PCR对141份腹泻犬粪便拭子样品中CCo V M基因片段(409 bp)进行了扩增,进一步对M基因进行了测序和分子进化性分析。结果表明:在141份样品中,38份样品为CCo V阳性(26.95%,38/141),其中CCo V-Ⅰ型和CCo V-Ⅱ型分别占13.16%(5/38)和86.84%(33/38)。M基因序列分析表明:5个CCo V-Ⅰ型毒株M基因序列的核苷酸同源性为95.7%~99.1%,推导的氨基酸序列同源性为97.4%~100%;33个CCo V-Ⅱ型毒株M基因序列的核苷酸同源性为97.4%~100%,推导的氨基酸序列同源性为100%;33个CCo V-Ⅱ型毒株与中国参考毒株HF3的核苷酸同源性为88.7%~96.2%,推导的氨基酸序列同源性为92.2%~97.4%。进化树分析表明:5个CCo V-Ⅰ型毒株与2个巴西参考毒株呈现出较近的亲缘关系;33个CCo V-Ⅱ型毒株形成五个亚群,与中国参考毒株(除NJ17毒株)和国外参考毒株表现出较远的亲缘关系。可见CCo V在犬腹泻中呈现出较高感染率,以CCo V-Ⅱ型毒株感染为主,并表现出遗传多样性。
