Research on Zedoary Turmeric Oil Injection Induction of Human Endometrial Carcinoma HEC-1-B Cell Apoptosis and Its Mechanism

摘要 目的:研究莪术油注射液对人子宫内膜癌HEC-1-B细胞体外增殖及相关蛋白表达情况的影响。方法:将体外培养的HEC-1-B细胞随机分5组,对照组、莪术油Ⅰ组、莪术油Ⅱ组、莪术油Ⅲ组、莪术油Ⅳ组。分别给与不同浓度的莪术油注射液培养48 h、72 h。采用MTT法检测HEC-1-B生长抑制率、Hoechest33258荧光染色观察HEC-1-B细胞核形态的变化、Western blot法检测相关蛋白的表达水平。结果:不同浓度莪术油注射液均对HEC-1-B细胞增殖具有抑制作用,同时诱导HEC-1-B细胞凋亡,上调Caspase-3蛋白的表达,下调Survivin蛋白的表达,其强度随着药物作用浓度及时间的增加而增强,呈时间-剂量依赖性,与对照组比较,差异有统计学意义(P<0.01)。结论:不同浓度莪术油注射液在一定浓度范围内可抑制HEC-1-B细胞生长,且诱导其凋亡,其机制可能与下调子宫内膜癌HEC-1-B细胞Survivin蛋白的表达,上调Caspase-3蛋白的表达有关。 Objective: To study the effect of zedoary turmeric oil injection on human endometrial carcinoma( HEC-1-B) cell proliferation and expression of related protein influence. Methods: HEC-1-B cells cultured in vitro were randomly divided into 5 groups,control group,Ⅰ,Ⅱ of zedoary turmeric oil of zedoary turmeric oil and zedoary turmeric oil Ⅲ,zedoary oil Ⅳ. They were treated with different concentrations of zedoary turmeric oil injection 48 h,72 h training. MTT assay was used to detect the growth inhibition rate of HEC-1-B,Hoechest33258 fluorescence staining to observe the changes of HEC-1-B cell nucleus,and the expression level of Western blot protein. Results: Different dose of zedoary turmeric oil injection had inhibitory effect on HEC-1-B cells,and at the same time,inducing HEC-1-B cell apoptosis,raising Caspase 3 protein expression,reducing Survivin protein expression,and its intensity increased with the increase of the concentration and time of the drug action,which showed a time-dose dependence,and the difference was statistically significant compared with the control group( P〈0. 01). Conclusion: Different concentrations of zedoary turmeric oil injection could inhibit the growth of HEC-1-B cells in a certain concentration range,and inducing its apoptosis,the mechanism may be related to the down-regulation of HEC-1-B expression of Survivin protein in endometrial carcinoma cells,the up-regulated expression of Caspase-3 protein.
