摘要
目的:分析痛风泰颗粒剂对Toll样受体基因TLR2、TLR3的调控作用,进而为解释该药物治疗痛风的作用机制提供理论依据。方法:首先痛风大鼠造模,获得原代炎性关节滑膜细胞。其次,将15只正常的SD大鼠随机分为生理盐水组,戴芬溶液组,低剂量、中剂量、高剂量痛风泰颗粒组,制备5种含药血清。最后,分别用上述5种血清刺激培养的炎性关节滑膜细胞,8 h后检测5组细胞TLR2和TLR3基因mRNA的表达。结果:经过不同条件的刺激后,TLR2基因只有中剂量痛风泰颗粒组的mRNA相对表达量与生理盐水组比较,具有显著差异(P<0.05),但总体趋势不明显;TLR3基因的mRNA相对表达量无任何趋势及表达差异(P>0.05)。结论:Toll样受体基因TLR2和TLR3可能并不是痛风泰颗粒剂治疗痛风的关键基因,更有价值的靶基因或调控因子有待于被发现。
Objective: To analyze the regulatory effect of pure Chinese herbal formula Tongfengtai Granule in the treatment of Tolllike receptor gene TLR2 and TLR3,and to provide theoretical basis for explaining the mechanism of the drug treatment of gout.Methods: First,gout rats to obtain primary inflammatory synovial cells. Secondly,15 normal SD rats were randomly divided into saline group,daffan solution group,low dose,middle dose,high dose of Tongfengtai granule group,prepared 5 kinds of medicated serum. Finally,the inflammatory joint synovial cells were cultured with the above 5 kinds of serum,After 8 hours,the expression of TLR2 and TLR3 gene mRNA in 5 groups were detected. Results: After stimulation with different conditions,TLR2 gene mRNA relative expression only in middle dose was significantly different from the normal saline group( P〈0. 05),but the overall trend was not significant; MRNA relative expression of TLR3 gene was not any trend and expression difference( P〉0. 05). Conclusion: Toll like receptor gene TLR2 and TLR3 may not be the key genes in the treatment of gout,the more valuable target genes or regulatory factors to be found.
