续断对兔膝骨关节炎模型滑膜中白介素-1β、骨桥蛋白和血管内皮生长因子影响的实验研究 被引量:4

Experimental study of dipsacus asperoids on interleukin-1β,osteopontin and vascular endothelial growth factor level in synovium of rabbit knee osteoarthritis models

摘要 目的探讨续断对兔膝骨关节炎(KOA)模型滑膜中白介素-1β(IL-1β)、骨桥蛋白(OPN)和血管内皮生长因子(VEGF)的影响。方法采用分层随机化的方法将动物分为5组,正常组、对照组、续断组低剂量、中剂量、高剂量组,每组各15只。对5组滑膜IL-1β、OPN和VEGF水平进行检测。结果治疗第8周5组滑膜IL-1β、OPN和VEGF浓度差异有统计学意义(F值分别为780.61、1 291.03、206.77,P<0.01);5组两两比较,对照组滑膜IL-1β、OPN和VEGF浓度显著高于低剂量组,而中剂量组显著低于低剂量组、高剂量组;正常组显著低于高剂量组,差异均有统计学意义(P<0.05)。治疗第8周续断组不同剂量与滑膜OPN和VEGF水平相关性研究显示,续断组滑膜IL-1β、OPN和VEGF水平与续断剂量均呈正相关(r值分别为0.98、0.99、0.90,P<0.01)。结论续断可降低KOA滑膜IL-1β、OPN和VEGF水平。 Objective To observe the effect of dipsacus asperoids on interleukin- 1β( IL- 1β),osteopontin( OPN) and vascular endothelial growth factor( VEGF) level in synovium of rabbit knee osteoarthritis( KOA) models. Methods The animal were divided into 5 groups,including normal group,control group and Radix dipsaci( high dose,medium dose,low dose)groups with stratified randomization,with 15 rabbit in each group. Interleukin- 1β,osteopontin and vascular endothelial growth factor level were detected in synovium in the five groups. Results On the eighth week after therapy,the concentration differences of interleukin- 1β,osteopontin and vascular endothelial growth factor level in synovium were statistically significant among the 5 groups( F = 780. 61,F = 1 291. 03,F = 206. 77,P〈 0. 05). Comparison was performed between each 2 groups in the 5 groups,the concentrations of synovial IL- 1β,OPN and VEGF in the control group were significantly higher than that in the low dose group,while that in middle dose group was significantly lower than that in low dose group and high dose group; the normal group was significantly lower than the high dose group,and the differences were statistically significant( P〈 0. 05). On the eighth week of treatment,the studies of correlation between different doses in the Dipsacus group and synovial OPN and VEGF levels showed that,in the Dipsacus group,synovial IL- 1β,OPN and VEGF levels were positively correlated with the dose of Dipsacus( r = 0. 98,r = 0. 99,r = 0. 90,P〈 0. 01). Conclusion Radix dipsaci can reduce the level of interleukin-1β,osteopontin and VEGF levels in synovium of KOA.
