摘要
目的研究DNA修复酶8-羟基鸟嘌呤DNA糖苷酶(OGG1)对PM2.5造成支气管上皮细胞线粒体损伤的保护作用及分子机制。方法 100 mg/L PM2.5分别染毒正常、5 mmol/L活性氧(ROS)清除剂N-乙酰-L-半胱氨酸(NAC)预处理1 h和OGG1过表达beas-2b细胞24 h,分别用Western blot、流式细胞术、Q-PCR检测线粒体OGG1表达、细胞内ROS水平、线粒体DNA拷贝数。结果 PM2.5抑制beas-2b细胞线粒体OGG1表达,线粒体中OGG1过表达抑制PM2.5所致细胞内ROS水平升高及线粒体DNA拷贝数降低。结论 OGG1抑制PM2.5诱导ROS升高、减轻线粒体损伤。
Objective To study the protective effect and molecular mechanism of OGG1 on PM2.5-induced mitochondrial damage in bronchial epithelial cells. Methods Normal, OGG1 overexpressed, and NAC pretreated Beas-2b cells were exposed to 100 mg/L PM2.5 for 24 h. The mitochondrial OGG1 expression, intracellular reactive oxygen species (ROS), and mito- chondrial DNA copies were detected using Western blot, flow cytometry, and Q-PCR, respectively. Results PM2.5 inhibited the mitochondrial OGG1 expression. Overexpression of mitochondrial OGG1 reduced PM2.5- induced ROS generation and mitochondrial DNA copies. Conclusion OGG1 can inhibit PM2.5- induced ROS production and alleviate DNA damage.
