乌司他丁对大鼠脑缺血再灌注后脑组织紧密连接蛋白1及血清神经元特异性烯醇化酶表达的影响 被引量:2

摘要 目的:探讨乌司他丁对缺血再灌注脑损伤的保护作用。方法:选取健康雄性SD大鼠72只,将所有大鼠以随机数字表法分为假手术组、缺血再灌注损伤模型组和乌司他丁处理组,每组各24只。建立大鼠缺血再灌注模型,检测3组大鼠脑组织紧密连接蛋白1(zonula occluden-1,ZO-1)及血清神经元特异性烯醇化酶(NSE)的表达变化。结果:与缺血再灌注损伤模型组大鼠相比,乌司他丁处理组大鼠脑组织中ZO-1蛋白表达显著增加,NSE表达明显减少,差异均有统计学意义(P<0.05)。结论:乌司他丁可通过抑制脑缺血再灌注后血-脑脊液屏障通透性的增加、减少NSE表达而发挥脑保护作用,其保护血-脑脊液屏障的机制可能与增加ZO-1蛋白的表达有关。 OBJECTIVE: To probe into the protective effects of ulinastatin on ischemia-reperfusion injury.METHODS: 72 healthy male SD rats were randomly selected to be divided into sham operation group,schemiareperfusion injury model group and ulinastatin group,with 24 cases in each. Schemia-reperfusion injury model was established,expression changes of zonula occluden-1( ZO-1) and neurone specific enolase( NSE) in brain tissues of three groups were detected. RESULTS: Compared with schemia-reperfusion injury model group,expression of ZO-1protein in ulinastatin group significantly increased,while NSE decreased significantly,with statistically significant difference( P〈0. 05). CONCLUSIONS: Ulinastatin can play a protective role in the brainm by inhibiting the increased cerebral ischemia and reperfusion brain barrier permeability,and reducing expression of NSE; and its protective bloodbrain barrier may be related to upregulation expression of ZO-1 protein.
