氧化苦参碱干预TGF-β1诱导的肺泡上皮细胞氧化应激反应和炎症因子表达作用研究 被引量:1

摘要 目的本研究旨在探讨氧化苦参碱(Oxymatnine,Oxy)对转化生长因子B。(TGF-β1)诱导肺泡上皮细胞氧化应激反应和炎症因子表达的干预作用。方法0.25、0.5、1g/L氧化苦参碱预处理A549细胞30min后,再给与5tzg/LTGF-β1共孵育培养48h,Westernblot检测诱导型一氧化氮合酶(iNOS)表达变化,试剂盒检测髓过氧化物酶(MPO)活性和丙二醛(MDA)水平,酶联免疫吸附测定(ELISA)法检测炎性因子肿瘤坏死因子a(TNF-a)和IL-6的表达。结果氧化苦参碱能有效干预TGF-β1诱导的A549细胞iNOS表达,降低MPO活性、MDA含量以及TNF-a、IL-6的水平。结论氧化苦参碱能有效抑制TGF-β1诱导的A549细胞氧化应激反应和炎症因子表达,氧化苦参碱可以作为抗肺纤维化治疗的候选药物。 Objective To investigate the effect of Oxymatnine (Oxy) on transforming growth factor 131 (TGF--β1 ) induced oxidative stress and inflammation factors expression in alveolar epithelial cells. Methods After treatment with 0.25,0.5,1 g/L Oxy separately for 30 min, A549 cells were co-incubated with TGF-β1(5μ g/L) for 48 h. The expression of iNOS was analyzed by Western blotting. The activities of myeloperoxidase (MPO) and the content of MDA were measured by spectrophotometry and the levels of tumor necrosis factor a (TNF-a) and interleukin-6 (IL-6) were tested by enzyme-linked immunosorbent assay (ELISA). Results Oxy could effectively decrease the expression of iNOS and reduce activities of myeloperoxidase (MPO), the content of MDA and the the levels of TNF-a and IL-6 induced by TGF-β1 . Conclusions Oxymatrine could inhibit TGF-β1 induced oxidative stress and inflammation factors expression in AS49 cells. Oxymatrine could be used as a new candidate drug for treatment of pulmonary fibrosis.
