脂联素rs266729多态性和肥胖在多囊卵巢综合征中交互作用研究 被引量:1

摘要 目的探讨脂联素rs266729多态性与多囊卵巢综合征(PCOS)关联性及其与肥胖交互作用。方法采用多聚酶链反应-限制性片段长度多态性(PCR-RFLP)方法分析2014年2月至2015年8月住院的120例PCOS患者和同期140例体检正常对照者的rs266729基因型。非条件Logistic分析各基因型与发病易感性的关系以及与肥胖的交互作用。结果携带C(CC/CT)基因个体患病风险较非C基因携带者(TT)风险明显增加1.55倍(OR=1.55,95%CI:1.41~3.71,P=0.00;校正OR=1.61,95%CI:1.51~4.01,P=0.00);非条件Logistic分析表明认携带CC/CT基因型肥胖个体PCOS罹患风险是携带TT基因非肥胖个体的3.88倍(OR=3.88,95%CI:2.72~5.76,P=0.00;RERI=2.51,95%CI:1.22~3.80;AP=0.54,95%CI:0.34~0.74;S=1.56,95%CI:1.12~2.83)。结论脂联素rs266729多态性增加多囊卵巢综合症罹患风险,且与肥胖存在多囊卵巢综合症发病中存在协同效应。 Objective To study the relationship between rs266729 polymorphism in endothelial adiponectin and polycystic ovarian syndrome( PCOS) and the interaction with obesity. Methods Genotypes of rs266729 of 120 hospitalized cases of PCOS and 140 controls selected from February 2014 to August 2015 were examined by polymerase chain reaction-restriction fragment length polymorphism( PCR-RFLP).The unconditional Logistic regression( ULR) was performed to assess the correlation of genotypes with susceptibility and the interaction with obesity. Results The risk of developing PCOS for CC / CT genotype carriers increased by 1. 55 times compared to TT( OR = 1. 55,95% CI: 1. 41- 3. 71,P = 0. 00; ORadjust= 1. 61,95% CI: 1. 51- 4. 01,P = 0. 00). ULR analysis showed that obesity-positive subjects with genotype CT / CC had 3. 88 times of PCOS risk of obesity-negative subjects with genotype TT( OR = 3. 88,95% CI: 2. 72- 5. 76,P =0. 00)( RERI = 2. 51,95% CI: 1. 22- 3. 80; API = 0. 54,95% CI: 0. 34- 0. 74; S = 1. 56,95% CI: 1. 12- 2. 83). Conclusion Adiponectin rs266729 polymorphism is associated with increased risk for PCOS,and its interaction with obesity has synergistic effects on risk of PCOS.
