摘要
目的探讨电针对帕金森病(PD)模型大鼠黑质超氧阴离子、超氧化物岐化酶的影响。方法选取雄性SPF级SD大鼠80只,随机将其分为正常组、溶剂对照组、模型组和治疗组,每组20只。正常组:不做任何处理。溶剂对照组:经大鼠背部皮下注射葵花油液,注射剂量为按每天2 mg/kg,每天注射一次,连续注射28 d。模型组:除连续28 d皮下注射鱼藤酮制备PD模型外,不再做其它特殊处理。治疗组:自大鼠造模一周后进行治疗。针灸取穴取风府、太冲两穴。比较各组大鼠的行为学改变、黑质的TH阳性细胞数,检测不同处理方法的抗超氧阴离子氧活性和超氧化物歧化酶活性。结果经电针治疗后的大鼠中,治疗组的行为学评分(6.44±1.21)显著低于模型组(7.31±1.46),差异有统计学意义(P<0.05)。治疗组大鼠的脑黑质TH阳性细胞数明显高于模型组(P<0.05)。模型组的抗超氧阴离子氧活性显著低于治疗组,差异具有统计学意义(P<0.05)。模型组和治疗组的超氧化物歧化酶活性均小于正常组(P<0.05),且治疗组显著高于模型组,差异具有统计学意义(P<0.05)。结论电针能通过提高抗超氧阴离子氧活性,增强抗氧化能力途径抑制神经元细胞凋亡,从而减轻PD模型大鼠的神经行为表现。
Objective To research the influence of electric acupuncture for superexide anion and superoxide dismutase in nigra of Parkin- song rat. Methods 80 male SPF SD rats were randomly divided into normal group, solvent control group, model group and treatment group, each group of 20 rats. Normal group : do none treatment. Solvent control group : sunflower oil was injected subcutaneously at the dose of 2 mg/kg once a day for 28 days. Model group : In addition to continuous subcutaneous injection of rotenone 28 d preparation of PD model, no other special treat- ment. Treatment group: one week after the model rats from the treatment. All the electric acupuncture in treatment group using "Fengfu" (GV16) and" Taiehong" (LR 3). Rats'behavior change and the number of TH positive neuron were compaTed between groups. Anti- supewxide activity and superoxide dismutase activity expression level was detected to compare the antioxidant capacity. Results After electroacupuncture treatment, the ethology score (6.44 ± 1.21 ) of the treatment group was lower than that of the model group (7.31 ± 1.46), the difference was statistically significant ( P 〈 0.05 ). After electreacupuncture treatment, the number of TH positive neuron of treatment group was significantly higher than that of the model group ( P 〈 0.05 ). The model group was significantly lower than the treatment group in anti - superoxide activity, with signifi- cant difference ( P 〈 0.05 ). The superexide dismutase activity of the model group and treatment group were less than that of the normal group ( P 〈 0.05 ), and the treatment group was significantly higher than that of model group, the difference was statistically significant ( P 〈 0.05 ). Conclusion Electric acupuncture can enhance anti - superexide activity and superoxide dismutase activity in nigra of Parkinson's rat, which may inhibit neuronal apoptosis, to alleviate the nerve behavior of the PD model rats.
