摘要
高分辨率熔解曲线技术是一种以饱和荧光染料实时监测双链DNA熔解过程、反映扩增子DNA序列特性的基因分析新技术,它具有特异、高效、快速、廉价、闭管操作等优点,目前已广泛应用于医学、生命科学、法医学、药材和食品鉴定等领域。本文对高分辨率熔解曲线技术及其在基因检测、突变扫描和荧光聚合酶链反应仪温度校验方面的应用进行综述。
High resolution melting (HRM) is new technology of genetic analysis which can monitor realtime change of dual-strand DNA melting by adding saturated fluorescent dyes to reflect the DNA sequence amplification characteristics. HRM has been widely applied in medicine, life science, forensic science, medicine and food identification and other fields because of its advantages of high specificity and efficiency, fast, cheap and close-tube operation. This article aims to review new advances of HRM and its applications in genetic testing, mutation scanning, and temperature calibration of polymerase chain reaction instrument.
