大豆GmAYI基因的克隆及初步功能预测

摘要 根据拟南芥At1G74730基因的保守序列设计引物,利用RT-PCR技术在大豆中克隆得到GmAYI基因。对GmAYI基因的结构域和启动子元件进行分析,结果表明:大豆GmAYI基因位于4号染色体上,含跨膜结构域,属于膜蛋白。该基因启动子区域含有激素相关应答元件和胚乳表达所必须的顺式作用元件。推测该基因可能参与调控大豆生长发育及外界环境胁迫应答等过程。进而对14个大豆品种中GmAYI基因的转录水平进行了定量分析,对GmAYI基因在14个大豆品种中的转录水平与单株粒数、单株粒重、百粒重、株高等产量相关性状进行相关性分析,结果表明:GmAYI基因的表达量与百粒重呈显著负相关,推测GmAYI可能是参与大豆产量相关性状调控的基因。 The primers were designed according to the conserved sequence of Arabidopsis Atl 674730 gene. The GmAYI gene was cloned from soybean by RT-PCR. The GmAY1 gene domain and the promoter element were analyzed. The results showed that the GmAY1 gene of soybean was located on chromosome 4, which contained transmembrane domain and belonged to mem- brane protein. The promoter region of the gene contained a hormone-related response element and a cis-acting element necessary for endosperm expression. The results suggested that the gene may be involved in the regulation of soybean growth, development and environmental stress response process. The transcript levels of GmAYI gene in 14 soybean euhivars were quantitatively analyzed. Based on the Correlation analysis of GmAYI gene transcripts in 14 soybean cuhivars with seeds number per plant, seeds weight per plant, 100-seed weight and plant height, the expression of GmAYI gene was negatively correlated with 100-seed weight, suggested that the GmAYI gene may be involved in the regulation of soybean yield-related traits.
