摘要
利用流式细胞法可以快速准确检测乳制品中的细菌总数,该方法的关键点是消除蛋白质、体细胞和脂肪的干扰。UHT乳的蛋白质水解条件:选择复合蛋白酶在60℃消化20 min。原料乳的蛋白质水解条件:选择0.6 mg/m L蛋白酶K在50℃消化20 min。离心脱脂的最佳条件为:离心转速9 000 r/min、时间8 min。
Arapid method of determination of total number of bacterial in dairy products by flow cytometry was investigated, the key point was how to eliminate the influence of milk proteins, somatic cells and fat. Protein hydrolysis conditions of Ultra- high pressure sterilization milk selected compound protease in 60 ℃ digesting 20 minutes. Protein hydrolysis conditions of raw milk selected 0.6 mg/mL protease K in 50 ℃ digesting 20 min. Centrifugal skim conditions were centrifugal speed 9 000 r/min and degreasing time 8 min.
