四神丸对溃疡性结肠炎大鼠结肠黏膜PPAR-γmRNA和蛋白表达的影响

The Influence of SiShen Pills on the Expressions of PPAR-γmRNA and Protein in Colonic Mucosa of the Rats with Ulcerative Colitis

目的:观察四神丸对三硝基苯磺酸(trinitrobenzene sulfonic acid,TNBS)/乙醇灌肠所致溃疡性结肠炎(ulcerative colitis,UC)模型大鼠结肠黏膜PPAR-γm RNA和蛋白表达的影响,探讨其作用机制。方法:将Wistar大鼠60只随机分为空白组、模型组、四神丸组(5 g/kg)及柳氮磺吡啶(SASP,0.3 g/kg)组,采用TNBS/乙醇灌肠法复制UC大鼠模型,灌胃给药后肉眼观察结肠大体形态损伤并评分,采用RT-PCR和免疫组化法检测PPAR-γm RNA和蛋白的表达。结果:模型组大鼠结肠组织黏膜层可见炎症和溃疡形成;与空白组比较,模型组大鼠结肠组织PPAR-γm RNA和蛋白表达量降低,差异有统计学意义(P<0.01);与模型组比较,四神丸组PPAR-γm RNA和蛋白的表达量升高,差异有统计学意义(P<0.01)。结论:四神丸对TNBS/乙醇法所致UC大鼠模型结肠黏膜PPAR-γm RNA和蛋白的表达有上调作用,提示四神丸治疗UC的作用机制可能与调控PPAR-γ相关信号转导途径有关。

Objective：To observe the effects of SiShen pills on the expressions of PPAR-γ mRNA and protein in colonic mucosa of TNBS/alcohol enema-induced ulcerative colitis（UC） rats,and explore its mechanism.Methods：Sixty rats were randomized into the blank group,the model group,SiShen pills group（5 g/kg） and SASP group（0.3 g/kg）,UC rat model was replicated by TNBS/alcohol enema,gross morphological injuries of the colon were observed under the naked eyes and scored,the expressions of PPAR-γ mRNA and protein were detected by RT-PCR and immunohistochemical method.Results：The inflammation and ulceration could be seen in the mucous layer of colon tissue of the rats in the model group;compared with the blank group,the expressions of PPAR-γ mRNA and protein decreased notably in the colon tissue of the rats in the model group,and the difference presented statistical meaning（P〈0.01）;compared with the model group,the expressions of PPAR-ymRNA and protein raised evidently in SiShen pills group,and the difference demonstrated statistical meaning（P〈0.01）.Conclusion：SiShen pills could up-regulate the expressions of PPAR-γ mRNA and protein in colon mucosa of UC rats,which suggests that the mechanism of SiShen pills in treating UC might be related to PPAR-γ-related signal transduction pathway regulation.
