摘要
目的构建人8-羟基鸟嘌呤DNA糖苷酶(h OGG1)左右臂载体敲除质粒对。方法针对OGG1基因,设计左臂识别序列3个,右臂识别序列2个,应用转录激活子样效应因子核酸酶技术(TALEN)一步法进行构建。结果成功将左右臂识别序列接入相应的表达载体中,构建出5个TALEN质粒,形成3X2组合模式。结论成功构建了敲除h OGG1基因的TALEN质粒对。
Objective To construct human 8-oxoguanine glycosylase-l(hOGG1) plasmid pairs with arm knock-out. Methods Five recognition sequences of left (3) and right arms (2) targeting for OGG1 gene were designed, and then assembled using transcription activator-like effector nuelease (TALEN) method. Results Five recognition sequences of left and right arms were transformed into expression vectors. Five TALEN plasmids were then constructed, with a 3×2 composite model. Conclusion TALEN plasmid pairs with hOGG1 gene knock-out are successfully constructed.
