NDRG2基因亚型对卵巢癌HO-8910细胞周期及凋亡的影响

Effects of N-myc downstream regulated gene 2 subtypes on cycle and apoptosis of ovarian cancer HO-8910 cells

摘要 目的构建表达N-myc下游调节基因2(NDRG2)两种亚型(NDRG2L,NDRG2S)的重组逆转录病毒载体,观察逆转录病毒介导的NDRG2基因对卵巢癌细胞HO-8910细胞周期及凋亡的影响。方法以携带人NDRG2基因的两种亚型逆转录病毒载体转染体外培养的卵巢癌细胞株HO-8910,采用Western-blot检测目的基因的表达,流式细胞术检测转染后对细胞周期和凋亡的影响。结果流式细胞仪检测显示,转染后G1期细胞明显减少,S期细胞明显增多(P<0.05),但转染后对细胞凋亡无明显影响(P>0.05)。结论通过逆转录病毒载体使细胞表达NDRG2基因,可以明显促进HO-8910细胞的增殖,但对细胞凋亡无明显影响。 Objective To construct NDRG2L and NDRG2S recombinant retroviral vectors, observe the effect of retrovirus-mediated NDRG2 gene on cell cycle and apoptosis of ovarian cancer HO-8910 cells. Methods NDRG2L and NDRG2S recombinant retroviral vectors were transfeeted into ovarian cancer HO-8910 cells, Western blot was used to detect the expression of target gene, flow eytometry was used to detect the effect on cell cycle and apoptosis. Results The results of flow cytometry showed that after transfection, the proportion of G1 phase cells decreased significantly, while the proportion of S phase cells increased significantly ( P〈0. 05 ) , but there was no significant impact on apoptosis of ovarian cancer HO-8910 cells ( P〉0. 05 ) . Conclusion The proliferation of ovarian cancer HO-8910 cells is significantly promoted by construction of retroviral vectors, but it has no significant impact on cell apoptosis.
