摘要
目的:研究胶质瘤组织中膜联蛋白A2(Annexin A2)和锌指转录因子Snail的表达及相应siRNA对胶质瘤细胞的调节作用。方法:收集脑胶质瘤组织和瘤旁组织,测定Annexin A2和Snail的表达量;培养脑胶质瘤细胞株U373-MG,转染Annexin A2、Snail、NC的siRNA后测定细胞活力、迁移及侵袭数目、增殖及上皮间质转化基因的表达量。结果:胶质瘤组织中Annexin A2和Snail的mRNA含量均显著高于瘤旁组织(P<0.05);Annexin A2-siRNA组的细胞活力以及Ras、Raf、MEK、ERK的mRNA含量均显著低于NC-siRNA组(P<0.05),迁移细胞数目、侵袭细胞数目以及E-cadherin、N-cadherin、Vimentin、α-SMA的mRNA含量与NC-siRNA组无显著性差异(P>0.05);Snail-siRNA组的迁移细胞数目、侵袭细胞数目以及N-cadherin、Vimentin、α-SMA的mRNA含量显著低于NC-siRNA组(P<0.05),E-cadherin的mRNA含量显著高于NC-siRNA组(P<0.05),细胞活力以及Ras、Raf、MEK、ERK的mRNA含量与NC-siRNA组无显著性差异(P>0.05)。结论:胶质瘤组织中Annexin A2和Snail的表达量显著增多,高表达的Annexin A2能够促进细胞增殖、高表达的Snail能够促进上皮间质转化。
Objective:To study the Annexin A2 and zinc finger transcription factor Snail expression in glioma tissue and the regulating effect of corresponding siRNA on glioma cells.Methods:Glioma and peri-tumor tissue were collected to determine Annexin A2 and Snail expression;glioma cell lines U373-MG were cultured and transfected with Annexin A2,Snail and negative control(NC)siRNA,and then the cell viability,number of migrating and invading cells as well as the expression levels of proliferation and epithelial-mesenchymal transition genes were detected.Results:Annexin A2 and Snail mRNA levels in glio-ma tissues were significantly higher than those in peri-tumor tissues;cell viability as well as Ras,Raf,MEK and ERK mRNA levels of Annexin A2-siRNA group was significantly lower than those of NC-siRNA group,and the migrating cell number and invading cell number as well as E-cadherin,N-cadherin,Vimentin andα-SMA mRNA levels were not significantly different from those of NC-siRNA group;migrating cell number and invading cell number as well as N-cadherin,Vimentin andα-SMA mRNA levels of Snail-siRNA group were significantly lower than those of NC-siRNA group,E-cadherin mRNA level was significantly higher than that of NC-siRNA group,and the cell viability as well as Ras,Raf,MEK and ERK mRNA levels were not significantly different from those of NC-siRNA group.Conclusion:Annexin A2 and Snail expression levels significantly increase in glioma tissues,highly expressed Annexin A2 can promote cell proliferation and highly expressed Snail can promote epithelial-mesenchymal transition.
