外源性硫化氢对人宫颈癌HeLa细胞的影响及作用机制 被引量:3

摘要 目的探讨外源性硫化氢对人宫颈癌Hela细胞的影响及其作用机制。方法将培养好的人宫颈癌HeLa细胞随机分为6组,正常对照组:正常培养,不作任何处理;Na HS组:加入400μmol/L的Na HS;Na HS+PDTC组:加入400μmol/L的Na HS和100μmol/L的PDTC(NF-κB通路抑制剂);Na HS+NS-398组:加入400μmol/L的Na HS和10μmol/L的NS-398(COX-2通路抑制剂);PDTC组:加入100μmol/L的PDTC;NS-398组:加入10μmol/L的NS-398。各组干预24 h,用CCK-8法测定细胞相对存活率,用ELISA法检测细胞培养上清液中血管内皮生长因子(VEGF)水平,用Transwell小室实验检测细胞迁移能力,用Western blotting法检测细胞内Caspase-1、Caspase-12、p-NF-κB p65和COX-2蛋白表达。结果干预24 h,与正常对照组、Na HS+PDTC组、Na HS+NS-398组比较,Na HS组细胞相对存活率、培养上清液VEGF水平、细胞迁移能力及NF-κB p65(除外Na HS+NS-398组)、COX-2蛋白表达升高,Caspase-1、Caspase-12蛋白表达降低(P均<0.05);PDTC组、NS-398组以上指标与正常对照组比较差异无统计学意义(P均>0.05)。结论外源性硫化氢可通过激活NF-κB/COX-2信号传导通路促进人宫颈癌HeLa细胞增殖、新生血管形成、细胞迁移。
