摘要
目的探讨金创抗菌膏对糖尿病大鼠溃疡创面愈合的影响及其作用机制。方法随机从50只大鼠中选出18只作为空白对照组,其余大鼠建立糖尿病模型。模型建造成功后,剔除造模失败的3只大鼠,随机将糖尿病大鼠分为金创抗菌膏组(n=15)和阴性对照组(n=14)。将三组大鼠进行皮肤溃疡模型建立后,金创抗菌组用金创抗菌膏换药,阴性对照组用生理盐水冲洗后包扎处理,对照组让其自然愈合。观察给药后大鼠的创面愈合情况。结果 1金创抗菌组大鼠在第7天和第14天的创面愈合率显著高于其他两组(P<0.05);2HE染色结果显示,与其他两组比较,金创抗菌组真皮层中炎细胞较少,纤维细胞数量减少呈长梭形整齐排列,再生复层扁平上皮细胞清晰;3苦味酸-天狼星红染色结果显示,与其他两组比较,金创抗菌组Ⅲ型胶原纤维明显较Ⅰ型胶原纤维多,且排列呈疏松的网状分布;4免疫组化结果显示,在第7天和14天时金创抗菌组MMP-13的平均光密度值均显著低于其他两组(P<0.05)。结论金创抗菌膏可能通过调节Ⅰ,Ⅲ胶原纤维分布和影响MMP-13表达治疗糖尿病溃疡。
Objective To investigate the effects of antiseptic ointment on the cutaneous ulcer healing in diabetic rats and the underlying mechanisms. Methods Eighteen rats randomly taken from 50 wistar rats served as con- trols, and the rest were used to establish the diabetic ulcer. After establishment of cutaneous ulcer, 3 were dropped because of failure of building models, the diabetic rats were randomly divided into treatment group treated with antiseptic ointment group (n = 15 ) and negative control group treated with saline (n = 14). After establishment of skin ulcers, antiseptic ointment group were treated with antiseptic ointment, negative control group treated with saline, and control group accepted no special treatment. Results ① Ulcers healed faster in the rats treated with antiseptic ointment than in both the blank and negative controls on the day 7, 14; ② HE staining showed less inflammatory infiltrate, reduced fibroblasts well arranged in fusiform, and re-epithelialization in antiseptic ointment group; ③ Pieric-sirius red staining showed more collagen Ⅲ and less collagen Ⅰ , which arranged in loose reticular pattern in antiseptic ointment group; ④ In compared with other two groups, the expression levels of MMP-13 decreased remarkably in antiseptic ointment group on the day 7, 14 (P 〈 0. 05 ). Conclusion Antiseptic ointment can accelerates ulcer healing via regulating the distribution of collagen Ⅰ and Ⅲ, and expression of matrix metalloproteinase 13 in diabetic rats.
