摘要
目的研究柚皮苷和柚皮素对ApoE^(-/-)小鼠主动脉粥样硬化斑块形成的作用。方法将ApoE^(-/-)小鼠分为模型对照组、柚皮苷和柚皮素不同剂量给药组,均给予高脂饲料。12周后检测小鼠血浆血脂和同型半胱氨酸水平,测定小鼠主动脉粥样斑块的大小及斑块中SR-A、LOX-1、ACAT1和ABCA1/G1表达。结果各组小鼠血脂水平无明显改变(P>0.05)。与其他组比较,大剂量柚皮苷和柚皮素给药组小鼠主动脉粥样斑块形成明显受到抑制(P<0.05);斑块中SRA、LOX-1和ACAT1表达下调(P<0.05),ABCA1/G1表达上调(P<0.05);血浆同型半胱氨酸水平明显下降(P<0.05)。结论大剂量柚皮苷和柚皮素可抑制ApoE^(-/-)小鼠主动脉粥样硬化斑块的形成。
Objective To explore the effect of naringin/naringenin on atherosclerotic plaque development of aorta in ApoE-/-mice. Methods ApoE-/- mice were randomly assigned to model control group, naringin or naringenin treatment groups.Each group was fed with high-fat diet.After 12 weeks, we examined the plasma level of lipid and homocysteine, the area of the atheroscleretic plaque and the expression of SR-A, LOX-1, ACATlandABCA1/GI in mice. Results There was no significant change in blood lipid levels between model control group and the naringin/naringenin treatment groups (P〉0.05).In contrast to other groups, the formation of atherosclerotic plaque in ApoE-/- mice was significantly inhibited innaringin/naringenin treatment groups (P〈0.05).The expression of SR-A, LOX-1 and ACATI were down-regulated (P〈0.05)while the expression of ABCA1/G1 was up-regulated ( P〈 0.05 ). The plasma level of homocysteine was reduced ( P 〈 0.05 ). Conclusion The formation of atherosclerotic plaque in ApoE-/- mice can be inhibited by high doses of naringin/naringenin.
