刮痧对腰椎间盘突出症模型大鼠神经损伤的保护作用 被引量:15

摘要 目的观察刮痧对腰椎间盘突出症模型大鼠背根神经节(DRG)组织形态学和P物质(SP)、神经生长因子(NGF)的影响,探讨刮痧对腰椎间盘突出症模型大鼠神经损伤的保护机制。方法 72只雄性SD大鼠随机分为假手术组、模型组、刮痧组,每组24只。采用自体髓核移植法建立腰椎间盘突出症大鼠模型。刮痧组于造模后第5天开始刮痧,假手术组及模型组不予干预措施。分别于造模后第4天(干预前)、干预后第6天、干预后第12天、干预后第18天(干预结束后)麻醉处死取材。应用HE染色法检测DRG形态变化,应用免疫组化法检测大鼠DRG中SP、NGF的含量。结果与假手术组相比,模型组的SP、NGF的表达维持在较高水平,差异具有统计学意义(P<0.01);不同治疗时间(干预后第6、12、18天),刮痧组的SP的表达水平与模型组相比明显降低(P<0.01),刮痧组的NGF的表达水平与模型组相比显著升高(P<0.01),且治疗时间越长,各指标变化越明显;HE染色结果显示,假手术组无明显的炎性水肿,刮痧组局部炎症反应及形态学改变较模型组明显减轻。结论刮痧能够有效减轻大鼠DRG的病理损伤程度,抑制炎性因子SP的表达,促进蛋白质NGF的增长,发挥神经保护作用。 OBJECTIVE To observe the effects of Gua Sha therapy on histomorphological changes,substance P(SP)and nerve growth factor(NGF)in dorsal root ganglion(DRG)of rat models,and to explore neuroprotective effects of Gua Sha therapy on lumbar disc herniation(LDH)model rats.METHODS A total of 72 male SD rats were divided into 3groups randomly:sham operation group,model group and Gua Sha group(n=24).The sham operation group were treated with sham operation;the rat model of LDH was established by using autologous nucleus pulpous.Gua Sha group were given Gua Sha therapy on the 5th day after the model establishment.Sham operation group and model group were given no therapy intervention.The ipsilateral L5 DRGs were removed to detect the morphological changes after HE staining and to measure the expression of SP and NGF by immune-histochemistry on the 4th day after operation(before intervention),the 6th day after intervention,the12 hday after intervention,and the 18 th day after intervention(at the end of intervention).RESULTS Compared with sham operation group,model group showed significantly increased expression of SP and NGF(P 〈0.01).In different treatment times(the 6th,12 th and 18 th days),Compared with the model group,the expression level of SP in DRG significantly decreased in the Gua Sha group(P〈0.01),while NGF significantly increased(P〈0.01).As the treatment time lasted,the index changes were the more obvious.HE staining results showed that no obvious inflammation reaction were found in sham operation group,and the immune-inflammatory reaction of DRG in Gua Sha group was obviously alleviated.CONCLUSION Gua Sha therapy can alleviate pathological damage of DRG in rats,inhibit the expression of inflammatory factor SP,promote protein expression of NGF and protect the nerve.
