摘要
目的 检测6-磷酸果糖激酶-2/果糖双磷酸酶-2同工酶3(PFKFB3)在胃癌组织中的表达,分析PFKFB3对胃癌细胞增殖和迁移能力的影响.方法 用实时定量反转录聚合酶链反应(RT-qPCR)方法检测87例胃癌患者癌和癌旁组织中PFKFB3 mRNA表达水平,并用统计学方法分析PFKFB3 mRNA表达水平与临床病理特征的相关性.用小干扰RNA (siRNA)干扰胃癌细胞MGC803中PFKFB3的表达,并用噻唑蓝(MTT)法比较干扰后胃癌细胞增殖能力的改变,Transwell法比较干扰后胃癌细胞迁移能力的改变.结果 87例胃癌患者癌组织中PFKFB3的mRNA水平平均是癌旁组织的2.1倍.胃癌组织中PFKFB3的高表达与脉管内癌栓形成、淋巴结转移和TNM分期明显相关.siRNA干扰胃癌细胞中PFKFB3的表达后,胃癌细胞的增殖能力和迁移能力的明显下降.结论 PFKFB3在胃癌癌发生发展过程中发挥重要作用.
Objective To investigate the expression of 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase-3 (PFKFB3) in human stomach cancer and its clinical significance as well as its effect on cell proliferation and migration.Methods The expression of PFKFB3 in human stomach cancer tissues was examined by real-time reverse transcriptase-polymerase chain reaction (RT-qPCR) analysis.The relationship of PFKFB3 expression with clinicopathological features of stomach cancer patients was analyzed by statistic analysis.Stomach cancer MGC803 cells were transfected by PFKFB3 small interfering RNA (siRNA).The migration and proliferation abiliies of stomach cancer cells were determined using transwell cell migration assay and methyl thiazol tetrazolium (MTT) cell proliferation assay,respectively.Results The results revealed that PFKFB3 mRNA level was significantly increased in tumor tissues compared to adjacent non-malignant tissues.Furthermore,correlation analysis indicated that PFKFB3 mRNA high expression was significantly associated with lymph node metastasis,intravascular cancer embolus,and TNM stage.Knockdown of PFKFB3 inhibits cell proliferation and migration ability in MGC803 cells.Conclusion PFKFB3 might paly an important role during the development of human stomach cancer.
