摘要
目的:获得一种呼吸道铜绿假单胞菌生物膜检测方法,探讨藻酸盐合成相关基因的表达对临床治疗的影响。方法:提取60例呼吸道铜绿假单胞菌感染患者气管内病变组织,实时荧光定量PCR检测藻酸盐合成相关基因algD、algB、algU、mucA的表达,60例患者根据是否耐药分组,分为耐药组(R组)40例,非耐药组(NR组)20例,根据治疗的难易程度,将60例患者分为治疗困难组(D组)38例,治疗容易组(E组)22例。并进行基因algD、algB、algU、mucA的表达与临床抗生素耐药及治疗难易之间的相关性分析。结果:经细菌培养证实的铜绿假单胞菌感染病例60例,呼吸道病变组织均有algD、algB、algU、mucA基因的表达;耐药组(R组)和非耐药组(NR组)对比,algD、algU表达有统计学差异,algB、mucA表达无统计学差异;容易组(E组)和困难组(D组)对比,algD、algU、algB、mucA差异无统计学意义;细菌耐药和治疗难易的相关性分析显示,细菌耐药和临床治疗难易相关。结论:algD与铜绿假单胞菌藻酸盐的合成有着直接的正相关性,可以作为判断是否有黏液型铜绿假单胞菌生长及生物膜形成的依据,而algU与algD的表达有正相关性。
Objective :To obtain a respiratory Pseudomonas aeruginosa biofilm detection methods ,explore the affect of the expression of the related alginate synthesis genes to the clinical treatment .Methods :Specimens were taken from the tracheal lesions of Pseudomonas aeruginosa infection cases ,real-time quantitative PCR was performed to detect the expression of alginate biosynthesis-related gene algD ,algB ,algU ,mucA .And correlation between gene algD ,algB ,algU ,mucA .Expression and clinical treatment effect and antibiotic resistance was statistically analysed .Results :For all of the bacterial culture confirmed Pseudomonas aeruginosa infection cases ,the alginate biosynthesis-related gene Expression was detected ;comparison of resistant group (R group) and non-resistant group (NR group) ,algD ,algU expression significantly difference ,algB ,mucA difference was not significant .Conclusion :Expression of algD and Pseudomonas aeruginosa alginate synthesis has a direct positive correlation .Detect of algD can be used to determine whether there is Pseudomonas aeruginosa biofilm formation .and the expression of algU have a positive correlation with algD .
