摘要
目的通过一测多评法同时测定野菊花Chrysanthemum indicum L.中5种有机酸的含有量。方法该植物70%乙醇提取液的分析采用Agela Venusil XBP C18色谱柱(250 mm×4.6 mm,5μm);流动相甲醇-0.2%磷酸,梯度洗脱;体积流量0.8 m L/min;检测波长326 nm;柱温20℃;进样量5μL。以绿原酸为内标,建立其他4种有机酸的相对校正因子。结果绿原酸、咖啡酸、3,4-二咖啡酰奎尼酸、3,5-二咖啡酰奎尼酸和4,5-二咖啡酰奎尼酸分别在0.007 92~0.158 4、0.000 368~0.007 36、0.010 2~0.204、0.010 08~0.201 6、0.009 92~0.198 4μg范围内线性关系良好,平均加样回收率(RSD)分别为98.26%(1.48%)、96.30%(2.26%)、98.35%(2.08%)、99.16%(1.84%)、98.26%(2.10%)。相对校正因子重复性良好,一测多评法与外标法所得结果相近。结论该方法简便快速,准确可靠,可用于野菊花的质量评价。
AIM To simultaneously determine the contents of five organic acids in Chrysanthemum indicum L.by quantitative analysis of multi-components by single maker( QAMS). METHODS The analysis of 70% ethanol extract of this plant was performed on a 20 ℃ thermostatic Agela Venusil XBP C18column( 250 mm × 4. 6 mm,5 μm),with the mobile phase comprising of methanol-0. 2% phosphoric acid flowing at 0. 8 m L / min in a gradient elution manner,the detection wavelength was set at 326 nm,and the injection volume was 5 μL. With chlorogenic acid as an internal standard, the relative correction factors of another four organic acids were established.RESULTS Chlorogenic acid,caffeic acid,3,4-O-dicaffeoylquinic acid,3,5-O-dicaffeoylquinic acid and 4,5-Odicaffeoylquinic acid showed good linear relationships within the ranges of 0. 007 92-0. 158 4 μg,0. 000 368-0. 007 36 μg,0. 010 2-0. 204 μg,0. 010 08-0. 201 6 μg and 0. 009 92-0. 198 4 μg,whose average recoveries( RSDs) were 98. 26%( 1. 48%),96. 30%( 2. 26%),98. 35%( 2. 08%),99. 16%( 1. 84%) and 98. 26%( 2. 10%),respectively. The reproducibilities of relative correction factors were good. The results obtained by QAMS and external standard method were similar. CONCLUSION This simple,accurate and reliable method can be used for the rapid quality control of C. indicum.
