参麦注射液体外诱导骨肉瘤MG-63细胞凋亡的机制研究 被引量:3

摘要 目的:研究参麦注射液体外对骨肉瘤MG-63细胞生长的影响及作用机制。方法:体外培养MG-63细胞,以不同浓度的参麦注射液作用于细胞,MTT法检测肿瘤细胞生长抑制率;流式细胞术测细胞凋亡率和细胞周期;ELISA法测凋亡基因Caspase-3、Caspase-9的含量;Bradford法测SOD、MDA含量。结果:参麦注射液能显著抑制MG-63细胞的增殖,呈剂量依赖性;增加MG-63细胞的凋亡率,并诱导出现S细胞周期阻滞;作用MG-63细胞后,Caspase-3和Caspase-9蛋白表达量增加;升高细胞内SOD的活性,降低MDA的含量。结论:参麦注射液抑制人骨肉瘤MG-63细胞增殖的作用机制与诱导细胞凋亡和将细胞周期阻滞于S期有关,其诱导凋亡的机制与Caspase家族,以及增加机体的抗氧化功能有关。 Objective: To investigate the effects and its mechanism of Shenmai injection on osteosarcoma MG-63 cells. Methods: MG-63 Cells were cultivated in vitro, given with different concentrations of Shenmai injection, the inhibitory rate of the cells was measured by MTT assay, cell apoptosis and cell cycle was examined by flow cytometry, the content of Caspase-3 and Caspase-9 were measured by ELISA, and the content of SOD, MDA were measured by Bradford. Results: Shenmai injection inhibited the growth of MG-63 cells in a dosedependent manner. The cell apoptosis rate was elevated and S phrase was arrested. Shenmai injection significantly increased the expressions of Caspase-3 and Caspase-9. And the activity of SOD increased, MDA decreased significantly. Conclusion: Shenmai injection inhibit the proliferation of MG-63 cells, its mechanism may be related to Caspase family, as well as increase the antioxidant function.
