摘要 目的:探讨脱嘌呤/脱嘧啶核酸内切酶1(Apurinic/apyrimidinic endonuclease-1,APE1)在糖尿病心肌病(diabetic cardiomyopathy,DCM)患者血清的表达改变和意义。方法:收集对照、糖尿病和DCM患者血清,ELISA法检测血清APE1的表达,FAM荧光标记法检测血清APE1的酶活性。结果:与对照组、糖尿病组相比,DCM组患者血清APE1的表达明显增加,而其DNA核酸内切酶活性显著降低。结论:表达增加而DNA修复功能减退的APE1可能参与糖尿病心肌病的发病过程。 Objective: To investigate the expression and significance of APE1 (Apurinic/apyrimidinic endonuclease-1 ) in diabetic cardiomyopathy ( DCM ) patients.Method: Levels of APE1 in serum were assayed by ELISA kit.Enzymic activity of serum APE1 in DCM patients was measured using FAM labeled fluorescence assay. Result: Serum levels of APE1 were dramatically elevated in DCM patients compared with the control group and the diabetes group, endonuclease activity of APE1 significantly lower than those of the control group and the diabetes group.Conclusion: Elevated serum APE1 with its DNA repair dysfunction might be involved in the pathogenesis of diabetic cardiomyopathy.
