摘要
目的:探讨红花注射液对大鼠离体心脏缺血/再灌注损伤的保护作用及其机制。方法:健康成年Wistar大鼠,通过Langendorff离体心脏灌流技术建立心肌缺血/再灌注模型并检测心肌血流动力学指标;检测心肌组织中超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)活性及丙二醛(MDA)含量;采用实时定量PCR及免疫印迹检测心肌组织PI3K、Akt、Sirt1表达变化。结果:红花注射液可明显恢复缺血/再灌注后大鼠离体心脏血流动力学病变;显著性升高机体心肌组织中SOD及GSH-Px活性,减少MDA含量;PI3K、Akt磷酸化水平及Sirt1蛋白表达明显增强。结论:红花注射液改善大鼠心肌缺血再灌注损伤的保护作用机制可能与其激活PI3K/Akt/Sirt1信号通路作用有关。
Objective:To investigate the protective effect and mechanism of Honghua injection on isolated rat hearts from ischemia/reperfusion injury.Method:healthy adult Wistar rats,by Langendorff from heart perfusion technique,were established myocardial ischemia/reperfusion model and detected of myocardial hemodynamic indexes.We detected the activity of SOD and GSH-Px and the content of MDA in myocardial tissue;and used real-time quantitative PCR and Western blot to detect the heart muscle tissue PI3 K,Akt,SIRT1 expression changes.Result:Honghua injection can significantly restore the ischemia/reperfusion and after reperfusion in rat isolated heart hemodynamic lesion;It significantly increased myocardial tissues in the activity of SOD and GSH-Px,and decrease the content of MDA.PI3 Kand Akt phosphorylation and SIRT1 protein expression increased significantly by Honghua injection.Conclusion:Honghua injection improve rat myocardial ischemia reperfusion injury protection mechanism,which may be related to the activation of PI3K/Akt/Sirt1 signaling pathway.
