四大品种菊花中不同成分含量比较 被引量:6

Comparison of Component Contents in Four Kinds of Flos Chrysanthemi

摘要 【目的】比较亳菊、贡菊、滁菊、杭菊等四大品种菊花中主要活性成分的含量。【方法】采用高效液相色谱法对菊花中的绿原酸、3,5-O-双咖啡酰基奎宁酸及木犀草苷进行含量测定。测定前2项指标的色谱条件:色谱柱为Diamonsil-C18柱(250 mm×4.6 mm,5μm),以乙腈为流动相A,以质量分数为0.4%的醋酸溶液为流动相B进行梯度洗脱,流速为1.0 m L·min^(-1),检测波长为330 nm,柱温30℃,进样量为10μL;测定木犀草苷的色谱条件:色谱柱为苯基硅烷键合硅胶为填充剂Agilent ZORBAX SB-phenyl(4.6 mm×250 mm,5μm),以乙腈—20 g/L磷酸溶液为流动相;流速为1.0 m L·min-1,检测波长为350 nm,柱温30℃,进样量5μL色谱。【结果】绿原酸、3,5-O-双咖啡酰基奎宁酸含量顺序为:亳菊>滁菊>杭菊>贡菊,含量差异有统计学意义(P<0.05);木犀草苷含量顺序为:滁菊>亳菊>贡菊>杭菊,含量差异均无统计学意义(P>0.05)。【结论】四大品种菊花中绿原酸、3,5-O-双咖啡酰基奎宁酸含量有明显差异,而木犀草苷含量差异不大。 Objective To compare the contents of main active components in four kinds of Flos Chrysanthemi, including Boju, Gongju, Chuju, and Hangju. Methods High performance liquid chromatography was applied for the determination of the contents of chlorogenic acid, 3,5-O-dicaffeoyl quinic acid and galuteolin in Flos Chrysanthemi. And the chromatography for the former indexes determination was performed on a Diamonsil-Cls column (250 mm ×4.6 mm, 5 μm) with an isocratic gradient elution using acetonitrile as the mobile phase A and a mass fraction of 0.4% of aqueous acetic acid solution as the mobile phase B. The flow rate was 1.0 mL·min- 1, the detection wavelength was set at 330 nm, the column temperature was 30 ℃, and the injection volume was 10 p,L; the chromatography for galuteolin determination was performed on an Aglient ZORBAX SB-phenyl column (4.6 mm × 250 mm, 5 μm) using acetonitrile 20 g·L-1phosphorie acid solution as the mobile phase. the flow rate was 1.0 mL·min-1, the detection warelength was set at 350 nm, the column temperature was 30 ℃, and the injection volumn was 5 p,L. Results The contents of chlorogenic acid and 3,5-O-dicaffeoyl quinic acid were in the decreasing sequence fromBoju, Chuju, Hangiu to Gongju, the differences being significan (P 〈 0.05). Galuteolin content was in the decreasing sequence from Chuju, Boju, Gongju to Hangju, the differences being not signifieant(P 〉 0.05). Conclusion The contents of ehlorogenie acid and 3,5-O-dicaffeoyl quinic acid vary greatly in 4 kinds of Flos Chrysanthemi, but the content of galuteolin is similar in the 4 kinds of Flos Chrysanthemi.
