双氢青蒿素对大鼠放射性肺炎的治疗作用

Study of Effects on the Treatment of Radiation Pneumonitis with Dihydroartemisinin in Rat

目的探讨双氢青蒿素对Wistar大鼠放射性肺炎(RP)的治疗作用。方法 60只雄性Wistar大鼠随机分成3组:空白组、模型组、双氢青蒿素(DHA)组。用6MV-X线直线加速器对大鼠双肺进行单次照射15Gy,照射后从第1天开始灌胃给药,连续28d,分别于照射后7d,14d,21d,28d处死。测定各组大鼠血液及肺泡灌洗液中红细胞和白细胞总数;大鼠肺脏组织学观察;ELISA法测血清中NF-κB,TNF-α和IL-6的表达。结果空白组大鼠肺表面光滑、无炎症反应;DHA组大鼠肺组织较模型组炎症反应轻,模型组肺组织炎症反应最明显。DHA组肺泡灌洗液中白细胞计数及血清中NF-κB,TNF-α和IL-6的表达均较模型组低,差异有统计学意义(P<0.05)。结论 DHA可通过降低细胞因子NF-κB,TNF-α和IL-6的表达水平,减轻炎症反应,起到治疗放射性肺炎的作用。

Objective To investigate the ability of dihydroartemisinin in treating radioactive pneumonia in rats. Methods Sixty male Wistar rats were randomly divided into three groups, blank control group, model group, dihydroartemisinin group. Except the blank con- trol group,the whole chest of rats was irradiated by 6MV X-rays for 15Gy dose once. The corresponding drug was given by gastric perfusion once a day from the first day on radiation to 28 days after radiation. After irradiation at 7d,14d,21d,28d,3 rats of each groups were killed. Determine the total number of red blood cells,white blood cells in the blood and the alveolar lavage fluid. Right lung was used to specimen observe and a portion of lung tissue was retained to observe the pathological changes by HE. Elisa method was used to check the expression of NF-KB,TNF-α,IL-6 in serum. Results There was no inflammatory response of the blank control group. The swelling appearance, flexibility and hemorrhage of lung surface of the dihydroartemisinin group were better than the model group. The inflammation of the model group was the most severe. The total number of white blood cells in the bronchoalveolar lavage fluid was significantly reduced. The dihydroartemisinin group and the blank control group compared with the model group,the expression of NF-KB,TNF-α, IL-6 were significantly reduced（ P 〈0.05 ）. Conclusion Dihydroartemisinin can remedy radioactive pneumonia in rats by inhibiting the expression of NF-KB,TNF-α,IL-6.
