摘要
目的研究肾衰营养胶囊对慢性肾衰竭(chronic renal failure,CRF)大鼠微炎症状态血管损伤的抗炎作用。方法 10只大鼠设为正常对照组,50只大鼠行5/6肾切除制作CRF模型,观察生长状态及检测肾功能,满足要求的随机分为模型组(9只)、中药治疗组(28只,低、中、高剂量)和氯沙坦钾治疗组(10只)。6周后,测定肾功能指标;采用ELISA法检测血清可溶性细胞间黏附分子1(s ICAM-1)、血管间黏附分子1(VCAM-1)水平;Western blot检测NF-κB p65、e NOS的水平;qRT-PCR法检测TNF-α、IL-6表达量。结果模型组及治疗组大鼠肾功能损伤指标(血Scr、BUN、血β2-微球蛋白、尿微量白蛋白)及血管炎症相关因子(s ICAM-1、VCAM-1、NF-κB p65、TNF-α、IL-6)水平显著高于正常对照组(P<0.01),而各治疗组明显低于模型组(P<0.05),中药中剂量组与氯沙坦钾组差异有统计学意义(P<0.01)。结论肾衰营养胶囊可减轻CRF大鼠肾功能损伤,抑制血管炎症相关因子的水平,改善CRF微炎症状态下血管损伤。
Objective To study the anti - inflammatory effects of Shenshuai Yingyang Capsule on blood vessel damage under micro inflammation in rats with chronic renal failure induced by 5/6 Nephrectomy. Methods Ten rats were assigned as normal control group, and 50 rats for establishment of 5/6 renal resection CRF model. The growth status and kidney function were recorded. The rats met criteria of renal failure were randomly divided into model group, capsule treatment groups (low, medium and high dose) and losartan potassium treatment group. Six weeks later, the renal func- tion indexes 'were tested. The serum soluble intercellular adhesion factor 1 ( slCAM 1 ) and vascular adhesion molecule 1 ( VCAM 1 ) levels were measured using ELISA method. The NF - kappa B p65 and eNOS levels were assessed using western blot. The TNF alpha and IL -6 expression were assessed using QRT PCR method. Results The rat kidney damage indexes (blood Scr, BUN, blood beta 2 -microglobulin, and urine trace albumin) and vascular inflammation related fac- tor ( sICAM 1, VCAM 1, the nf - kappa B p65, TNF alpha, and IL - 6) were significantly higher in model group and treatment group than those in normal control group (P 〈 0.01 ). Those indexes were all significantly lower in all treatment groups than those in model group ( P 〈 0.05 ) ; moreover it was more convincing in medium dose group and losartan potassi- um treatment group (P 〈 0.01 ). Conclusion Shenshuai Yingyang Capsule is demonstrated to reduce the CRF rats renal injury, inhibit inflammation related factor levels and improve the vascular injury under CRF micro inflammatory condition.
