三七总皂苷联合雷公藤多苷对胶原诱导关节炎大鼠血管内皮生长因子表达的影响 被引量:5

Effect of Panax Notoginsenosidum Combined with Tripterygium Glycosides on VEGF Expression in Rats with Collagen-induced Arthritis

摘要 目的观察三七总皂苷(PNS)联合雷公藤多苷(TG)对胶原诱导关节炎(CIA)大鼠血清血管内皮生长因子(VEGF)水平和在胸主动脉、心肌中表达的影响。方法将造模成功的CIA大鼠随机分为模型、TG(10 mg/kg·d)、PNS(200 mg/kg·d)+TG(10 mg/kg·d)和甲氨蝶呤(MTX,1.6 mg/kg·周)组,另设正常大鼠为正常对照组。给药6周后采血,开胸取心脏和胸主动脉。采用液相蛋白芯片技术检测血清VEGF含量,免疫组化法检测胸主动脉、心肌VEGF蛋白表达。结果与正常对照组比较,CIA模型组胸主动脉存在粥样硬化病变,胸主动脉及心肌可见炎性细胞浸润、脂质沉积,血清VEGF水平明显升高(P<0.05),胸主动脉及心肌VEGF蛋白表达显著上调(P<0.05);PNS+TG组可明显降低血清中VEGF水平、下调VEGF蛋白表达,与TG组和MTX组相比,差异均具有统计学意义(P<0.05)。结论中药有效成分TG与PNS配伍可明显改善CIA大鼠胸主动脉及心肌病变,其机制可能与下调血清及心肌、胸主动脉VEGF表达,从而发挥抗炎作用有关。 Objective To observe effect of Panax Notoginsenosidum(PNS) combined with Tripterygium Glycosides(TG) on serum levels of vascular endothelial growth factor(VEGF) and expressions in thoracic aorta and myocardium of rats with collagen-induced arthritis(CIA).Methods Successful CIA rat models were randomly divided into model,TG(10 mg / kg·d),PNS(200 mg / kg·d) + TG(10 mg / kg·d) and Methotrexate(MTX)(1.6 mg / kg·w)groups(n = 8 for each group),and normal rats were selected as control group(n = 8).Blood samples were collected after 6 weeks of treatment,and heart and thoracic aorta were obtained.Liquid phase of protein chip technology was used to measure serum VEGF levels,and VEGF protein expressions in thoracic aorta and myocardium were detected by immunohistochemical method.Results Compared with those in control group,rats in model group had thoracic aorta atherosclerosis,and inflammatory cells infiltration and lipid deposition were found in thoracic aorta and myocardium,and serum VEGF levels were significantly increased(P〈0.05),VEGF protein expressions in thoracic aorta and myocardium were significantly increased(P〈0.05); TG + PNS significantly reduced serum VEGF levels and down-regulated VEGF protein expressions,and its effect was significantly better than those in MTX and TG groups(P〈0.05).Conclusion Chinese medicine effective ingredient TG + PNS can significantly improve thoracic aorta and cardiac lesions in CIA rats,and its mechanism may be achieved by down-regulating VEGF expressions in serum,myocardium and thoracic aorta to play an anti-inflammatory effect.
