摘要
目的探讨PTEN(phosphatase and tensin homolog deleted on chromosome ten)与TGF-β(transforming growth factor beta)在前列腺癌中的表达及其临床意义。方法选取不同TNM分期的前列腺癌石蜡组织共32例,正常前列腺组织8例,通过Real-Time PCR对组织中PTEN和TGF-β的mRNA水平进行相对定量,比较二者在不同TNM分期的前列腺癌中表达水平;通过免疫组织染色对上述前列腺组织中PTEN和TGF-β蛋白的进行定位和阳性率分析。结果在T1、T2期前列腺癌组织中PTEN和TGF-β的mRNA水平与蛋白阳性率与正常组织比较差异无统计学意义(P<0.05);在T3、T4期的前列腺癌组织中,PTEN的mRNA水平与蛋白阳性率较正常组织和T1、T2期显著提高(P<0.01),TGF-β则明显下调(P<0.01),二者呈负相关(P<0.05)。结论 PTEN和TGF-β与前列腺癌的发展及恶性程度有关,与中晚期的肿瘤的增殖与迁移关系密切,二者的表达水平可辅助鉴定前列腺癌的分化程度与分期。
Objective To investigate expressions of phosphatase and tensin homolog does on chromosome ten(PTEN) and transforming growth factor beta(TGF-β) in prostate cancer and its clinical significance.Methods A total of 32 samples of paraffin-embeded prostate cancer tissues in different TNM stages and 8 samples of normal tissues of prostate cancer patients were selected,and mRNA related quantification levels of PTEN and TGF-β in tissues were detected by real-time PCR(polymerase chain reaction),and differences of the levels in patients with different TNM stages were compared.Location and positive rates of PTEN and TGF-β in different TNM stages were analyzed by immunohistochemical staining.Results There were no significant differences in mRNA levels and positive rates of PTEN and TGF-βin T1 and T2between prostate cancer and normal tissues.(P〈0.05).Values of mRNA levels and positive rates of PTEN were significantly increased(P〈0.01),while TGF-β mRNA levels of prostate cancer tissues in T3 and T4stages were significantly decreased compared with those in normal and prostate cancer tissues in T1 and T2stages(P〈0.01).Negative correlation was found between PTEN and TGF-β expressions(P〈0.05).Conclusion Expressions of PTEN and TGF-βare correlated with the development and malignant degree of prostate cancer.Meanwhile,it is closely correlated to the proliferation and migration of middle-late tumor,and the expressions can be use to adjuvant evaluation of differentiation and stages for prostate cancer.
