摘要
目的探讨巴豆醛对肺泡液体清除率的影响,并探讨其潜在的作用机制。方法应用考马斯亮蓝测定小牛血清白蛋白浓度的方法,评估小鼠在体肺泡液体清除率;采用蛋白质印迹法实验研究巴豆醛对细胞外信号调节蛋白激酶(ERK)1/2蛋白磷酸化的影响以及其对肺泡上皮钠通道表达的影响;用活性氧检测试剂盒检测巴豆醛对细胞内活性氧含量的影响。结果巴豆醛能够降低小鼠肺泡液体清除率,蛋白质印迹法结果显示其能够促进ERK1/2的磷酸化,抑制上皮钠通道亚基的表达,并且呈剂量依赖性提高细胞内的活性氧水平。结论巴豆醛能够抑制肺上皮细胞的液体清除,该抑制作用可能与其增加ERK1/2蛋白磷酸化,抑制上皮钠通道蛋白表达以及增加细胞内的活性氧水平有关。
Objective To explore the effects of crotonaldehyde on the alveolar fluid clearance (AFC) and to study the potential mechanisms. Methods AFC was measured using bovine serum albumin protein assays by Xylene brilliant cyanin G method. Western blotting was used to analyze the expression of epithelial sodium channels (ENaC) and extracellular signal-regulated protein kinases1/2 (ERK1/2) phosphorylation protein levels. Reactive oxygen species (ROS) Kit was used to detect the effect of crotonaldehyde on ROS levels. Results Crotonaldehyde could decrease the rate of AFC in mice/n vivo . Western blotting analysis showed that crotonaldehyde could increase the level of ERK1/2 phosphorylation and inhibit the expression of ENaC, meanwhile, it could elevate the ROS levels in H441 cells. Conclusion These data demonstrate that crotonaldehyde can reduce the alveolar ion transport, possibly related to the enhancement of ERK1/2 phosphorylation, inhibition of ENaC protein expression and increment of ROS level in H441 cells.
