HPLC法测定熊胆汁中牛磺熊去氧胆酸含量的研究

Study on Taurocholate Ursodeoxycholic Acid Content in Bear Bile by HPLC

目的建立熊胆汁中牛磺熊去氧胆酸含量的HPLC测定方法。方法用甲醇前处理熊胆汁;以十八烷基键合硅胶为填料;甲醇-磷酸二氢钠溶液(0.03 mol/L)(68:32)为流动相,并用磷酸调节p H值为4.4;柱温40℃;流速1.0 m L/min;进检测波长为210 nm。结果牛磺熊去氧胆酸进样浓度在0.1045 mg/m L^1.045 mg/m L之间呈良好的线性关系,r=0.9999,回收率在98%~102%之间,回收率RSD<1.5%。结论方法准确、简便、快速,可为熊胆汁的检测及监控提供技术依据和支持。

Objective： To establish a HPLC method to determine taurocholate ursodeoxycholic acid content in bear bile. Methods： Bear bile was pre - treated with methanol. Octadecyl bonded silica was taken as a filler, methanol - sodium dihydrogen phosphate solution （0. 03 mo]./L） （68：32） as mobile phase, and phosphoric acid was used to adjust to pH 4. 4. The column temperature was 40℃, with 1.0 mL/min of flow rate, 210 nm of detection wavelength. Results ： The sample concentration of taurocholate ursodeoxycholic acid showed a good linear relationship between 0. 1045 mg/mL and 1. 045 mg/mL, r = 0. 9999, with the recovery rate between 98% to 102% and recov- ery rate RSD 〈 1.5%. Conclusion： The method is accurate, simple and rapid, and can provide a technical base and support for the detec- tion and monitoring of bear bile.
