没食子酸对肺癌A549细胞的毒性作用及机制研究

摘要 观察没食子酸对肺癌A549细胞的毒性作用。将肺癌A549细胞分为对照组、没食子酸组(GA)、没食子酸+过氧化氢酶组(GA+CAT)和没食子酸+超氧化物歧化酶组(GA+SOD),通过检测细胞存活率、细胞形态变化、超氧阴离子(O_2^(·-))自由基和过氧化氢(H_2O_2)含量探讨作用机制。检测结果显示,GA组A549细胞存活率明显低于对照组,呈现死亡的A549细胞数量、H_2O_2和O_2^(·-)含量多于对照组(P<0.05)。GA-CAT组A549细胞存活率明显高于GA组,呈现死亡的A549细胞数量、H_2O_2和O_2^(·-)含量明显低于GA组(P<0.05)。GA-SOD组A549细胞存活率明显低于GA组和GA-CAT组,H_2O_2含量明显高于GA组和GA-CAT组,O_2^(·-)含量明显低于GA组(P<0.05)。GA-SOD组O_2^(·-)含量只略低于GA-CAT组(P>0.05)。GA诱导H_2O_2和O_2^(·-)大量生成可能是GA降低肺癌A549细胞存活率的重要作用机制。 To investigate the toxic effects of gallic acid on lung cancer A549 cells. Lung cancer A549 cells were divided into control group,gallic acid group( GA group),gallic acid- catalase group( GA- CAT group) and gallic acid-superoxide dismutase group( GA- SOD group). The cell survival rate,the cell morphological changes,the levels of super oxygen anion( O2·-) free radicals and the contents of hydrogen peroxide( H2O2) were analyzed to clarify the mechanisms. The results showed that the survival rate of A549 cells in GA group was lower than that in control group,the number of dying cells,the contents of H2O2 and O2·-were higher than those in control group( P 〈 0. 05). The survival rate of A549 cells in GA- CAT group was higher than that in GA group,the number of dying cells,the contents of H2O2 and O2·-were lower than those in GA group( P 〈 0. 05). The survival rate of A549 cells in GA- SOD group were lower than that in GA group and GA- CAT group,but the content of H2O2 in GA- SOD group was higher( P 〈0. 05). The content of O2·-in GA- SOD group was lower than that in GA group( P 〈 0. 05). Compared with GA- CAT group,the content of O2·-in GA- SOD group decrease slightly( P 〉 0. 05). The possible mechanisms of A549 cell survival rate reduction were H2O2 and O2·-production that they were triggered by GA.
