叶片山海绵水溶性活性产物的大孔吸附树脂分离及其抗肿瘤活性

The Enrichment Product from Sponge Mycale phyllophila Water Extract by a Macroporous Resin and Its Anti-tumor Activity

摘要 海绵拥有丰富的生物活性产物,以叶片山海绵(Mycale phyllophila)为材料,建立了以DA201-C大孔吸附树脂为核心的从水提液中提取活性产物的方法.以Lowry法跟踪得率,此方法对小分子肽类的回收率约为57%;粗提物的主要成分为肽类,约占89%.细胞活性检测表明粗提物对C6神经胶质瘤细胞、A2780-CP卵巢癌顺铂耐药细胞和HepG2肝癌细胞具有明显的抑制作用.本工作为深入研究叶片山海绵水溶性活性产物中的抗肿瘤组分奠定了基础. Marine sponges contain abundant bioactive compounds.This study established a method that a DA201-C macroporous ad-sorption resin was used to enrich bioactive products from Mycale phyllophila water extract.Lowry's assay showed that this method recovered about 57% of the total small molecular peptides from the water extract.The majority compounds of the enrichment product are peptides,accounting for about 89%.In addition,the enriched product has obvious inhibitory activities to C6 glioma cells, A2780-CP ovarian cancer cisplatin-resistant cells and HepG2 hepatoma cells.This study lays the foundation for in-depth study on the anti-tumor compound from M.phyllophila water extract.
