砧木种类对化橘红黄酮类成分含量的影响 被引量:3

摘要 目的对采用不同种类的砧木培育出的化橘红药材进行黄酮类成分含量测定,初步探究砧木种类对其黄酮类成分含量的影响。方法以柚皮苷为对照品,利用紫外分光光度法测定各样品中总黄酮含量,以HPLC法同时测定各样品中柚皮苷及野漆树苷含量。结果所建立的可见分光光度法的加样回收率为95.81%,RSD为1.06%。HPLC法同时测定柚皮苷与野漆树苷含量在进样量为1.54-30.80μg和0.07-1.40μg范围内与色谱峰面积积分值呈现良好的线性关系,相关系数分别为0.998,0.997。柚皮苷与野漆树苷的加样回收率分别为98.11%(RSD=3.42%)、98.63%(RSD=2.37%)。不同砧木培育的化橘红黄酮类成分含量差异显著。与凤尾化橘红相比,本地柚砧木凤尾野漆树苷含量与其相近,其他品种均明显低于凤尾化橘红。结论不同种类的砧木对化橘红中黄酮类成分的含量影响较大。非本地柚为砧木芽接后化橘红药材野漆树苷含量显著下降,以本地柚为砧木嫁接凤尾芽苗培育出的化橘红品质较优。 Objective To study the content of flavonoids from Exocarpium Citri Grandis cultivated from different kinds of rootstocks,and to preliminarily investigate the effects of rootstocks on flavonoids content of Exocarpium Citri Grandis.Methods Using naringin as control sample,we tested the total flavonoids content in each sample by ultraviolet spectrophotometry,and established a HPLC method for the simultaneous determination of naringin and sumac glycoside content in the samples.Results The spectrophotometric method had the recovery rate of 95.81 %,and RSD=1.06 %.The results of HPLC method for simultaneous determination of naringin and rhoifolin contents showed that good linear relationship was obtained in the range of 1.54-30.80 μg(naringin)and 0.07-1.40 μg(rhoifolin),and the recovery of naringin and rhoifolin was 98.11 %(RSD=3.42 %),98.63 %(RSD=2.37 %)respectively.The contents of flavonoids of Exocarpium Citri Grandis cultivated from different rootstocks varied significantly.Rhoifolin content of phoenix shooting graft +local pomelo rootstock group was similar to that of the normal phoenix shooting graft group,but the content of other rootstock groups was much lower.Conclusion Rootstocks show obvious effect on flavonoids content of Exocarpium Citri Grandis cultivated from different kinds of rootstocks.Rhoifolin content of Exocarpium Citri Grandis cultivated from phoenix shooting graft and local pomelo rootstock is the highest,showing the best quality.
