摘要
目的建立新西兰兔血浆中利多卡因浓度的高效液相色谱(HPLC)测定方法。方法采用Symmetry Shield RP18(5μm,4.6×250mm)色谱柱,流动相为乙腈-0.01mol·L^(-1)磷酸盐缓冲液(12∶88),检测波长为210nm。结果该方法日内、日间精密度RSD均小于5%,样品回收率在86%~105%,利多卡因在1.13μg·m L^(-1)~113μg·m L^(-1)范围内线性关系良好(r=0.9999)。结论该方法专属性好,灵敏度高,适用于新西兰兔血浆中利多卡因的含量测定。
OBJECTIVE To establish a HPLC method for determination of lidocaine in New Zealand rabbit plasma. METHODS The column Symmetry Shield RP18 (5 μm ,4. 6 - 250mm) was utilized in the experiment. The mobile phase consisted of acetonitrile and 0. 01mol . L-1 phosphate buffer (12:88). The detective wavelength was 210nm. RESULTS The within-day and day-to-day RSD were less than 5% ,the recovery rate ranged from 86% to 105% ,and good linearity was obtained in the range of 1. 13μg . mL-1 - 113μg .mL-1 with good correlation( r = 0. 9999). CONCLUSION This method provided a sensitive and accurate way in the determination of lidocaine in New Zealand rabbit plasma.
