Exendin4拮抗Aβ1-42所致大鼠识别记忆损伤的保护效应研究

摘要 目的探讨Exendin4拮抗淀粉样β蛋白(amyloidβ-protein,Aβ)引起大鼠识别记忆功能伤害的保护作用。方法将80只健康成年雄性SD大鼠随机分为对照组(PBS组)、Aβ1-42组、不同剂量Exendin4组(0.02、0.20、2.00 nmol)、不同剂量Exendin4(0.02、0.20、2.00 nmol)联合Aβ1-42组,每组10只。各组大鼠经侧脑室给药后进行新物体识别实验,旷场中间隔5 h分别放置2种物体,每次大鼠自由探索10 min。检测指标为新物体识别指数(NOI)=探索新物体时间/(探索新物体时间+探索旧物体时间)×100%。结果 Aβ1-42组大鼠NOI明显低于对照组,0.20、2.00 nmol Exendin4联合Aβ1-42组大鼠NOI明显高于Aβ1-42组,差异均有统计学意义(P<0.05)。结论 Exendin4预处理能剂量依赖性对抗Aβ1-42引起的大鼠识别记忆损伤。 Objective To investigated the protective action of Exendin4 for antagonizing recognition memory impairment induced by amyloidβ-protein-42(Aβ1-42) in rats. Methods Eighty healthy adult male Wistar rats were randomly divided into the control group(PBS),Aβ1-42,group,different concentrations of Exendin4(0.02,0.20,2.00 nmol)groups and different concen-trations of Exendin4(0.02,0.20,2.00 nmol)combined Aβ1-42 group,10 cases in each group. The rats in all groups were performed the new object recognition experiment after lateral cerebral ventricular medication injection. Two kinds of object were respectively placed in the open-field for interval of 5 h. The each time free exploration in rat lasted for 10 min. The detection index was the novel object recognition index(NOI)=exploring novel object time/(exploring novel objects time+exploring familiar objects time) ×100%. Results The NOI percentage in the Aβ1-42 group was significantly lower than that in the control group(P〈0.01) and the NOI percentage in the 0.20,2.00 nmol Exendin4 combined Aβ1-42 group was significantly higher than that in the Aβ1-42 group,the differences were statistically significant (P〉0.05). Conclusion Exendin4 pretreatment can dose-dependently antago-nize against the Aβ1-42-induced recognition memory impairment in rats.
