摘要
目的探讨NKX2.2在尤因肉瘤(Ewing’s sarcoma,ES)中的表达及与其它小圆细胞肿瘤的鉴别诊断价值。方法采用免疫组化En Vision两步法检测49例ES组织和43例其它小圆细胞恶性肿瘤中NKX2.2、CD99及FLI1的表达,分析NKX2.2并联合其它指标在ES诊断中的敏感性和特异性。结果 NKX2.2在ES中的阳性率为71.43%(35/49),在其它小圆细胞肿瘤中阳性率为9.30%(4/43);NKX2.2在ES中表达的敏感性和特异性分别为71.43%和90.70%。CD99在ES中表达的敏感性和特异性分别为100%和23.26%,FLI1在ES中表达的敏感性和特异性分别为87.76%和30.23%。联合检测NKX2.2与CD99及FLI1对ES诊断的敏感性和特异性分别为65.31%和100%。结论 NKX2.2蛋白是ES诊断和鉴别诊断中有用的标志物,联合检测三者有助于ES与其它小圆细胞肿瘤的鉴别。
Purpose To investigate the expression of NKX2. 2 in Ewing' s sarcoma ( ES), as well as the differential diagnosis value with other small round cell tumors. Methods Immunohistochemical of EnVision two-step staining was applied to detect the expression of NKX2. 2, CD99 and FLI1 in 49 ES cases and 43 other small round cell malignant tumor cases. Then, the sensitivity and specificity of NKX2. 2 in ES with or without combining with other biomarkers were also evaluated. Results The positive expression rate of NKX2. 2 in ES was 71.43% (35/49), while the rate was 9. 30% (4/43) in the other small round cell tumors. The sensitivity and specificity of NKX2. 2, CD99 and FLI1 in discrimination between ES and other small round cell tumors were 71.43% , 90.70% vs 100%, 23.26% and 87.76%, 30. 23%, respectively. Furthermore, the sensitivity and specificity of combined expression of NKX2. 2, CD99 and FLI1 in ES was 65.31%, 100%, respectively. Conclusion NKX2. 2 protein is a useful marker for ES diagnosis and differential diagnosis with other small round cell tumors, especially when combined with CD99 and FLI1.
