摘要
目的:观察苦参素对实验性自身免疫性脑脊髓炎(EAE)大鼠模型中JAK2/STAT3表达的影响。方法:用豚鼠全脊髓匀浆和完全弗氏佐剂制成的抗原乳剂复制EAE模型,将40只Wistar大鼠随机分为正常组、模型组、苦参素预防组和治疗组,观察记录大鼠的临床症状,苏木素-伊红(HE)染色,勒克司坚牢蓝(LFB)染色和皮尔苏斯基氏(Bielschowsky)染色观察病理改变,RTPCR法检测脊髓中JAK2的表达、ELISA法测定EAE大鼠脑中STAT3含量的变化。结果:与模型组相比,苦参素有效延迟EAE大鼠的发病时间,明显降低临床神经功能学评分(P<0.01),改善EAE大鼠中枢神经系统炎症浸润(P<0.01)、髓鞘脱失(P<0.01)和轴索损伤(P<0.01)程度,下调脊髓中JAK2和脑组织中STAT3的表达(P<0.01,P<0.01),且预防组和治疗组之间的差异性具有统计学意义(P<0.05)。结论:苦参素对EAE大鼠有防治作用,其作用机制可能与下调JAK2/STAT3的表达有关。
OBJECTIVE To observe effects of matrine on expression of JAK2/STAT3 in rats with experimental autoimmune encephalomyelitis (EAE). METItODS Antigen emulsion made by homogenate of guinea pig spinal cord and complete Freund's adjuvant was used to copy EAE model, and 40 Wistar rats were randomly divided into normal group, vehicle group, matrine pre- vention group and therapy group. Clinical scores were observed and recorded. Hematoxylin and eosin (HE), Luxol fast blue (LFB) and Bielsehowsky staining were used to observe pathological changes. RT-PCR and ELISA were used to detect expres- sion of JAK2 in spinal cord and change of STAT3 in brain of EAE rats. RESULTS Compared with model group, matrine de- layed onset of EAE and reduced EAE clinical scores (P〈0. 01), and also alleviated inflammatory infiltration (P〈0. 01), demy- elination (P〈0. 01) and axonal injury (P〈0. 01) in central nervous system of EAE rats. Moreover, expression of JAK2 in spi- nal cords and levels of STAT3 in brain were obviously reduced (P〈0. 01), and the difference between prevention and therapy groups was significant (P〈0. 05). CONCLUSION Matrine has protective effects on EAE rats, its mechanism may be related to down-regulation of expression of JAK2/STAT3.
