重组解脂耶氏酵母全细胞催化合成反10,顺12-共轭亚油酸的条件优化

Optimization of production of trans-10,cis-12-conjugated linoleic acid by whole-cell catalysis using recombinant Yarrowia lipolytica

摘要 采用重组解脂耶氏酵母全细胞催化合成反10,顺12-共轭亚油酸(t10,c12-CLA)单体,为了提高t10,c12-CLA的产量,对全细胞催化条件进行优化。通过含有油酸的培养基摇瓶培养36 h获得菌体细胞,经反复冻融处理制备全细胞催化剂。优化后的催化体系条件为:温度28℃,磷酸盐缓冲液(p H 7)浓度0.1 mol/L,转速200 r/min,无需限制氧气。在优化后的反应条件下催化反应40 h,t10,c12-CLA产量达到15.6 g/L,转化率为62.2%。 Trans - 10, c/s - 12 - conjugated linoleic acid( tlO, c12 - CLA) was produced by whole - cell catalysis using recombinant Yarrowia lipolytica. In order to increase the productivity of t10, c12 - CLA, the catalysis conditions were optimized. Whole - cell catalyst was prepared by cultivating in medium con- taining oleic acid for 36 h and treating with repeated freeze and thaw. The optimal catalysis system condi- tions were obtained as follows: temperature 28 ℃, phosphate buffer (pH 7 ) concentration 0.1 mol/L, speed 200 r/min, without oxygen limit. And the yield of t10, c12 - CLA was 15.6 g/L after reaction for 40 h under the optimal conditions, with a conversion rate of 62.2%.
