斯氏油脂酵母细胞内油脂含量的快速检测方法研究

Rapid determination of oil content in Lipomyces Starkeyi cells

以斯氏油脂酵母为研究对象,探讨了利用苏丹黑B染色定量测定其胞内油脂含量的可行性,对苏丹黑B染色法测定胞内油脂含量的条件进行了优化。结果表明:斯氏油脂酵母经苏丹黑B染色处理后在600 nm处有最大吸收峰;在沸水浴中加热6 min和不添加盐酸的情况下,斯氏油脂酵母苏丹黑B染色效果最佳;建立了斯氏油脂酵母胞内油脂含量与吸光值A600的线性回归方程,线性关系显著(R2=0.992 9)。该方法与酸热-有机溶剂法测定结果相对误差在允许范围内,可用于快速测定斯氏油脂酵母胞内油脂含量。

With the Lipomyces Starkeyi as study object, the feasibility of quantitative determination of oil content in its cells by Sudan black B staining method was investigated and the determination conditions were optimized. The results showed that Lipomyces Starkeyi stained with Sudan black B had the maximum absorbance at 600 nm. Under the conditions of heating in boiling water bath for 6 min without hydrochloric acid, the effect of staining Lipomyces Starkeyi with Sudan black B was the best. The linear regression equation of oil content in Lipomyces Starkeyi cells and absorbance A600 was established, and the linear re- lationship was significant（R^2 = 0. 992 9）. The relative error between the Sudan black B staining method and acid heat -organic solvent was in the allowable range, so the Sudan black B staining method could be used to rapidly determine the oil content in Lipomyces Starkeyi ceils.
