摘要
目的分析余姚市手足口病的病原谱构成,了解EV71的基因特征。方法收集1 827份手足口病病例标本,用实时荧光PCR检测并分型,样品进行肠道病毒71型(EV71)VP1基因的扩增和核苷酸序列测定,并用DNAstar和Blast软件对测序结果进行比对分析。结果 2010年-2013年1 827份标本中EV71和Cox A16的检出率分别为37.71%、24.25%,其他肠道病毒的检出率为22.28%;EV71 VP1蛋白氨基酸序列相似性为98.65%~100.00%,11株EV71基因分型属于C4a基因亚型。结论 2010年-2012年余姚市手足口病流行的主要病原体为EV71和Cox A16,2013年以其他肠道病毒为主。流行的EV71属C4a基因亚型,并具有高度相似性。
Objective To understand the constitution of pathogen that cause hand- foot- mouth disease( HFMD),and to research the gene characteristics of EV71. Methods Stool specimens of HFMD patients were collected and detected with real-time fluorescence polymerase chain reaction. Samples were conducted for EV71 VP1 gene amplification and nucleotide sequence determination,and the sequencing results were compared and analyzed in DNAstar and Blast software. Results 1 827 specimens were collected from 2010 to 2013,the detection rates of EV71 and Cox A16 were 37. 71% and 24. 25%,and the detection rate of other enterovirus was 22. 28%; the similarity rate of EV71 VP1 protein amino acid sequence was within 98. 65%-100. 00%. The genotyping of 11 strains of EV71 belonged to C4 a gene subtype. Conclusion The dominant pathogen of HFMD in Yuyao were EV71 and Cox A16 from 2010 to 2012,and other enterovirus were in high prevalence in 2013. The prevalent subtype of EV71 belonged to C4 a gene subtype with high similarity.
