兖州卷柏提取物对CCl_4致小鼠肝损伤的保护作用 被引量:3

Protective Effect of Ethanol Extract from Selaginella Involvens on Carbon Tetrachloride-Induced Hepatic Damage in Mice

摘要 目的研究兖州卷柏提取物对四氯化碳(CCl_4)引起肝损伤小鼠的保护作用。方法 48只KM小鼠随机分成6组。Ⅰ组和Ⅱ组为正常对照组和肝损伤模型组,分别每天一次性灌胃20mL/kg生理盐水;Ⅲ组每天一次性灌胃80mg/kg水飞蓟素;Ⅳ-Ⅵ组分别每天一次性灌胃40,80,120mg/kg兖州卷柏提取物。各组连续给药7d。末次给药2h后,除正常对照组外,其他各组一次性腹腔注射10mL/kg的0.2%CCL_4。禁食不禁水18h后,称质量;摘眼球取血,分别测定血清中谷丙转氨酶(ALT)和谷草转氨酶(AST)活性;颈椎脱臼法处死小鼠,取小鼠肝脏并称质量,另取小鼠肝脏右叶的相同部位,用10%甲醛固定,进行石蜡切片和H-E染色,检测肝组织病理学变化;取肝脏组织匀浆,测定总抗氧化(T-AOC)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)活性。结果兖州卷柏提取物各剂量组ALT和AST活性均显著低于模型组(P〈0.01),且趋于正常小鼠水平;与模型组比较,兖州卷柏提取物各剂量组均能显著提高肝脏T-AOC和CAT活性(P〈0.01)。除低剂量组外,其他实验组与模型组之间SOD活性均存在显著差异(P〈0.01);兖州卷柏提取物能减轻肝组织损伤程度。结论兖州卷柏提取物对CCl_4致小鼠急性肝损伤具有保护作用。 involvens (EES were randomly group and mod I) Objective To evaluate the hepatoprotective effect of ethanol extract from Selaginella on carbon tetrachloride (CC14)-induced hepatic damaged mice. Methods The animals vided into six groups, and treated as follows: Group I and Group I] were normal control group, and they were both given 20 mL/kg normal saline. Group Ill were given 80 mg/kg silymarin. Group IV, V and YI were given EESI with the dose of 40, 80, 120 mg/kg respec- tively. All groups were given medicines or normal saline via intragastric administration once a day lasting for 7 days. 2 h after the last intragastric administration, all animals were given 0.2 % CC14 with the dose of 10 mL/kg except normal control group. After 18 h, all animals were weighted and blood sample were collected by eye bleeding method. The activities of alanine transaminase (ALT) and aspartate transami- nase(AST) of blood sample were evaluated. Following the sacrifice for all the animals, the livers were dissected. Thereafter, the activities of total antioxidant capacity (T-AOC), catalase(CAT) and superox- ide dismutase(SOD) of liver supernatant were tested. The livers were fixed in 10% formaldehyde at room temperature for histological analysis. Results Administration of EESI significantly reduced the level of ALT, AST(P〈0.01). Compared with model group, administration of EESI could significantly enhance the level of T-AOC, CAT(P〈0.01). Groups other than the low dose group, significant differences in SOD activity existed between model group and EESI administration groups(P〈0.01). Administration of EESI could alleviate liver injury in sinusoidal spaces in CC14-induced hepatic mice. Conclusion EESI may protect hepatic injury in CC14-induced mice.
