摘要
目的:利用实时定量逆转录-聚合酶链反应(RT-PCR)技术来研究丹参酮Ⅰ对人胃癌MGC-803细胞中cmcy、p53、bcl-2、bax基因表达的影响,初步阐明丹参酮Ⅰ抑制胃癌细胞生长的作用机理。方法:将人胃癌细胞MGC-803接种于6孔板中培养24h,利用试剂盒提取细胞总RNA,通过逆转录方式获得稳定的DNA,采用RT-PCR法来检测丹参酮Ⅰ对细胞cmcy、p53、bcl-2、bax基因的差异性表达情况。结果:丹参酮Ⅰ显著下调了cmyc、bcl-2的基因表达,显著上调了p53、bax基因表达。结论:丹参酮Ⅰ通过下调cmyc、bcl-2以及上调p53bax基因表达,促进细胞凋亡,抑制人胃癌MGC-803肿瘤细胞的生长。
Objective : To investigate the mechanism of Tanshinone I for the inhibition of MGC - 803 gastric cancer cells by real time RT -PCR. Methods :The human gastric cancer MGC -803 cells were cultured in 6 -well plates for 24 hours, and then the mRNA was extracted for the study'of the gene expression of crecy, p53, bcl - 2 and bax by RT - PCR. Results : Thegene expression of cmcy and bcl - 2 were significantly down - regulated and the gene expressions of p53 and bax were significantly up - regulated by tanshinone I. Conclusion : TanshinoneI can promote the apoptosis and inhibit the growth of human gastric cancer MGC - 803 cell via the up - regulation of p53 and bax and down - regulation of cmey and bcl - 2 gene expression.
