摘要
目的:观察心衰康及其拆方对充血性心力衰竭(CHF)大鼠尿液水通道蛋白2(AQP2)的影响,并评价心衰康及其拆方不同用药周期对心衰大鼠水液代谢作用机制及差异。方法:对大鼠行冠状动脉结扎术造成心梗后心衰模型,按超声评价的左室射血分值分为6组,分别为模型组、心衰康全方组、益气组、温阳组、活血组、利水组。连续灌药4周及8周后观察动物尿量,并采用实时定量PCR法检测心衰康及其拆方对心衰大鼠尿液AQP2的影响。结果:心衰大鼠进行药物干预4周后,大鼠尿液中AQP2的含量较模型组比较均降低,其中心衰康全方组作用最明显,利水组次之,且利水组较其他拆方组比较均有差异(P<0.05);而药物干预8周后,心衰康全方组的效果同样最为明显,而利水组效果与益气、温阳、活血组类似(P>0.05)。结论:短期应用心衰康全方及利水中药能够降低心衰大鼠尿液AQP2的表达,而益气、温阳、活血中药对心衰大鼠尿液AQP2无明显影响。随着用药时间的延长,益气、温阳、活血药对AQP2的作用逐渐增强,而利水药作用无明显改变。
Objective:To observe Xinshuaikang and its disassembled recipes on heart failure congestive heart failure (CHF) rats' aquaporin - 2 ( AQP2 ) and evaluate the effect on water metabolism in rats with heart failure. Methods : Rats which underwent coronary ligation model of heart failure after myocardial infarction caused by ultrasound evaluation of left ventricular ejection fraction were divided into six groups ,namely the model group ,Xinshuaikang whole formula group, Oi -benefiting group, Yang -war- ming group, blood circulation - activating group, urine - promoting group. Continuous perfusion for 4 weeks, we observed urine and used real - time quantitative PCR to detect the effect of Xinshuaikang and its disassembled recipe on heart failure rats' AQP~. Re- sults : After intervention for 4 weeks, AQP2 in the urine of rats with heart failure were reduced compared with that of the model group. The whole formula group had the most obvious effect, followed by urine - promoting group. And the comparisons between u- fine - promoting group with the other disassembled recipe groups were all had difference( P 〈 0.05 ). However ,8 weeks after in- tervention, the whole formula group also had the best effect. And the effect of urine - promoting group was similar as that of the other disassembled recipes groups. Conclusion : Short - term use of Xinshuaikang and its disassembled recipes can reduce the expression of AQP2 of heart failure rats and the drugs for benefiting Qi,warming Yang and activating blood circulation had no effect on AQP2. As the time prolonging, the effect of the drugs for benefiting Qi, warming Yang and activating blood circulation would be enhanced but the drugs for promoting urine has no obvious change.
