摘要
目的:研究明日叶查尔酮(AC)对小鼠H22肝癌微血管密度(MVD)表达水平和癌细胞增殖活性的影响。方法:将50只皮下接种肝癌H22细胞株的小鼠随机分为5组,每组10只。AC低、高剂量组分别每日经口灌胃5mg/kg和50mg/kg AC,恩度(Endostar)组(阳性对照组)给予等量生理盐水灌胃并腹腔注射4mg/kg恩度,空白对照组及肿瘤对照组则分别给予等量的生理盐水灌胃及腹腔注射,连续注射10d后处死小鼠,测定瘤重、体重和抑瘤率,取瘤组织,用免疫组化法检测各组小鼠肝癌组织MVD的表达水平,并用四甲基偶氮唑蓝(MTT)法检测各组小鼠肝癌细胞增殖活性。结果:高剂量AC组与肿瘤对照组肝癌组织MVD计数分别为(4.5±1.286)与(9.9±1.175),差异有统计学意义(P<0.05);高剂量AC组与肿瘤对照组的肝癌细胞增殖活性吸光度分别为(0.506±0.032)和(1.330±0.089),差异有统计学意义(P<0.05)。结论:AC可降低小鼠H22肝癌MVD表达水平及肿瘤细胞增殖活性水平,对小鼠H22肝癌组织新血管生成具有明显的抑制作用。
Objective: To study effects of angelica keiskei chalcone on MVD expression of H22 in tumor-bearing mice. Methods: 50 mice were inoculated with H22 cells and divided into 5 groups, 10 cases in each. The AC low dose group and high dose group were administered with AC, 5mg/kg and 50 mg/kg daliy respectively. The endostar group (the positive control group) was given endostar 4 mg/ kg by intraperitoneal injection; and the tumor control group and the blank control group were given normal saline. All mice were sacrificed after 10 days. The tumor weight, body weight, and tumor inhibition rate were determined. The protein expression level of MVD was assessd by immunohistochemistry method, and the proliferation activity of H22 cells was determined by MTT assay. Results: The average MVD counts were (9.9± 1.175), (4.5±1.286) in the AC high dose group and the tumor control group respectively, with a statistically significant difference. The cell proliferation levels were (0.982 ±0.052) and (0.506 ±0.032) in the AC high dose group and the tumor control group respectively, with a statistically significant difference. Conclusion: AC can reduce the MVD expression and the activity of tumor cell proliferation and effectively inhibit tumor angiogenesis H22 in tumor-bearing mice.
