玉米低聚肽和姜黄素对小鼠的醒酒功能研究 被引量:12

摘要 评价玉米低聚肽和姜黄素单独使用及联合应用时对醉酒小鼠的醒酒活性,探讨其作用机理。将清洁级雄性KM种小鼠随机分为4组,即模型对照组和低、中、高剂量组。各剂量组灌胃相应受试物,玉米低聚肽模型对照组灌胃等量双蒸水,姜黄素模型对照组灌胃含5.6%乙醇的双蒸水。0.5h后每组分别灌胃白酒(56°)。灌胃结束后立即测定行为学指标(翻正反射消失时间、醉酒持续时间、攀附时间),30 min后测定血液中乙醇、乙醛浓度,并测定肝组织乙醇脱氢酶(ADH)、乙醛脱氢酶(ALDH)、细胞色素P450(CYP450)、过氧化氢酶(CAT)、黄嘌呤氧化酶(XOD)和超氧化物歧化酶(SOD)活性。与模型对照组相比,玉米低聚肽能延长小鼠攀附时间和翻正反射消失时间,缩短醉酒时间,降低血液中乙醇和乙醛含量。高剂量姜黄素可明显改善小鼠醉酒后的行为学指标,一定程度上降低血中乙醇和乙醛浓度。玉米低聚肽和姜黄素均可增强小鼠肝组织ADH和ALDH活性,提高CYP450、CAT和SOD含量,降低XOD活性。玉米低聚肽和姜黄素联合应用时对醉酒小鼠的活性强于单独使用姜黄素,但与单独使用玉米低聚肽组无显著差异。在该实验条件下,玉米低聚肽和高剂量姜黄素对醉酒小鼠具有明显的醒酒活性。 The antialcoholic effects of corn oligopeptides (COP) and curcumin used alone or in combination on drunk mice were evaluated and its corresponding antialcoholism was investigated. Male KM mice were randomly divid- ed into 4 groups, i.e. the model control group, the low-dose group, the middle-dose group and the high-dose group. Each dose group was garaged with corresponding test sample. The model control group of COP was gavaged with the same volume of distilled water while the model control group of curcumin was gavaged with the same volume of dis- tilled water containing 5.6% ethanol. After 0.5 h, dose group and model control group were gavaged with the optimal dose of dye liquor (56~). The behavioral indicators (loss of righting reflex time, drunk duration and cling time) were determined immediately. The concentrations of ethanol and acetaldehyde in blood, and the activity of ADH, ALDH, CYP450, CAT and XOD in liver were measured after 30rain. Compared with the model control group, climbing time and disappearance time of righting reflex of mice in the COP groups were extended and drunk time was shortened.. Meanwhile, the blood levels of ethanol and acetaldehyde were lower in the COP treated groups compared with those in the model control group. High-dose curcumin could obviously improve the behavioral indicators of drunk mice and lower the blood levels of ethanol and acetaldehyde to some degree. Both in COP treated groups and curcumin treated groups, the activities of ADH and ALDH, and the contents of CYP450, CAT and SOD in liver were higher than those in the model control group while the activity of XOD was lower than that in the model control group. The antialcoholic ability of drunk mice treated by COP and curcumin used in combination was stronger compared to curcumin used a- lone, while there was no obvious difference between the group treated by COP and curcumin used in combination and COP treated groups. The COP and high-dose curcumin can improve the antialcoholic ability of drunk mice under this experimental circumstance.
