摘要
目的从废弃羽毛堆积处土壤中分离筛选具有高效降解羽毛角蛋白的细菌。方法形态学观察、生化测定和16 Sr DNA序列分析鉴定菌株,测定蛋白水解研究酶活性检及酶特性。结果鉴定该菌为嗜麦芽窄食单胞菌,降解羽毛的最适发酵培养温度为30℃,最适p H是7.0,Fe^(2+)对酶活有促进作用,Cu^(2+)、Zn^(2+)、Mn^(2+)对酶活有明显抑制作用。结论嗜麦芽窄食单胞菌角蛋白酶的作用温度和p H宽泛、稳定性好,具有良好的开发价值和应用前景。
Objective:To isolate and characterize feather-degrading bacteria from soil of discarded feathers. Methods:Morphological observation,biochemical analysis and 16SrDNA sequence analysis were used for identifying bacteria,and the enzyme activity and enzyme properties were studied by mensurating protein hydrolysis. Results:The strain was identified as a strain of the genus Pseudomonas,The optimal pH and temperature for growth were 7. 0 and 30 °C,Fe2 + promote the ac-tivity of the enzyme,Cu2 + ,Zn2 + ,Mn2 + inhibited the activity of the enzyme. Conclusion:This bacteria has the potential for the application of feather keratin sources because the keratinase of Stenotrophomonas maltophilia functional temperature and pH are broad and stable.
