人肺腺癌A549细胞株胰岛素受体与放射敏感性的实验研究

摘要 目的研究人肺腺癌A549细胞放射治疗组与对照组的胰岛素受体(IR)的表达,探讨胰岛素受体与放射敏感性的关系。方法将随机分为对照组(0 Gy)和放射治疗组(剂量为2 Gy、4 Gy、6 Gy、8 Gy、10 Gy、12 Gy 6 MV X线)人肺腺癌A549细胞进行放射治疗,放疗后培养25天后用免疫组化S-P法测定肺腺癌A549细胞株各标本的胰岛素受体表达情况。结果胰岛素受体(IR)主要分布于A549细胞膜上。小剂量(2 Gy、4 Gy)组与对照组(0Gy)胰岛素受体表达强阳性率差异无统计学意义。6 Gy、8 Gy、10 Gy、12 Gy放射治疗组与对照组相比,胰岛素受体强阳性率与对照组差异有统计学意义(P<0.005),其中12 Gy剂量组强阳性率最高。结论胰岛素受体与放射敏感性负相关,ISR强阳性的肿瘤细胞对射线抗拒。胰岛素受体有可能成为预测肺癌放射敏感性的分子标志物。 Objective:To find relation between insulin receptorand cancer radiosensitivity. Methods:A549 cells were randomly divided into two groups ,matched group(0 Gy)and radiation groups(2 Gy,4 Gy,6 Gy,8 Gy,10 Gy,12 Gy with 6MV X rays). The cells were radiated and the cells were cultured for 25 days after radiotherapy and expression of lung adenocarcinoma A549 cell line samples were detected by Immunohistochemistry(SP method ). Results:Insulin receptor was located mainly in the A549 cell membrane. There was no difference in the expression of insulin receptorbetween small dose(2 GY,4 GY)groups and control group(0 GY). The groups with the doses of 6 GY,8 GY,10 GY and 12 GY were compared with the control group,and the insulin receptor expression had statistical significance( P 〈 0. 005),and the highest dose group with dose of 12 GY had strongest positive expression. Conclusion:Insulin receptor and radiosensitivity have negative correlation,and tumor cells with ISR strong positive are resistant to radiation. Insulin receptor may become one of cancer radiosensitivity predictive molecular markers.
